Objiective:To investigate the effect of sesamin on cholesterol and serum matrix metalloproteinase-2 and matrix metalloproteinase-9 (MMP-2 and MMP-9) in atherosclerosis rabbits, and explore the effection of sesamin prevent atherosclerosis.Methods:1. Group:30 Japanese white rabbits were used.Rabbits fed normal diet were used as control group (n=10), and that fed high-choleserol diet as atherosclerotic model group (n=10).The remained rabbits (n=10) were fed the high cholesterol diet containing sesamin (4mg/d, by stomach irrigating).2. Model:Bovine serum albumin (250mg/kg) were used once by ear-edge intravenous injection to these rabbits fed high cholesterol diet at beginning of this study.3. Methods:The aortic fragments were cut and stained with hematoxylin to odserved the pathologic chang.Oxidase method was used to determine concentrations of total cholesterol (TC), triglycerides (TG), high-density lipoprotein (HDL) and low-density lipoprotein (LDL) at the time of 0,5,8th week-end. Immunohistochemistry method was used to examine Aortic wall MMP-2 and MMP-9 of each group at the time of 8th week-end. ELISA method was used to determine serum MMP-2 and MMP-9 of each group at the time of 8th week-end.Results: (1) The TC,TG,LDL in sesamin guorp were significantly lower than that in model group (P<0.01).(2) The pathologic change of arteria aorta endomembrane in sesamin group was singnificanty lighter than that in model group.(3) Immunohistochemistrymethod:To observethe expression of MMP-2on aortic wall; Model group aortic express positive cells was significantly higher than the normal controlgroup; Sesamin groupwas higher than the normal control group, significantly lower than the model group.(4) Serum MMP-2 and MMP-9 in sesamin group was significantly lower than that in model group. Serum MMP-2 and MMP-9 in model group was significantly higher than that in normal control group(P<0.05).Conclusions:(1) Sesamin dffectively decreased the concentrations of TC,TG and LDL,reduced the lipid depositin area,which may relate to the antioxidatie effection as well as the inhibition of ox-LDL synthesis.(2) Sesamin effectively decreased the serum MMP-2 and MMP-9 in the atherosclerotic plaque.Sesamin could inhibit inflammation.(3) Sesamin can reduce foam cells and macrophages to express MMP-2 and MMP-9 through lowering blood lipids, inhibiting the inflammatory response.(4) Sesamin can effectively on inhibiting atherosclerosis.Sesamin is potential to be one of green health food for economi primary prevention from atherosclerosis.
