Objective:To find a new molecular markers for differential diagnosis of human B-cell lymphoma, and to set theoretical basis to be used for molecular targeted therapy to human B cell lymphoma, we detect the expression of Transcribed Ultraconserved Regions(T-UCRs)in B-cell lymphoma of human and mice.Then compare the expression level with that of B cell in human normal lymph node and in spleen or bone marrow of mice respectively. And investigate the correlation between the expression and that of the human B cell lymphoma.Then explain their possible function in the initiation and progression of B-cell lymphoma.Methods:Screen the T-UCRs which are express higher in B cell lymphoma cells of mice by gene Microarray.Extract total RNA from the normal B cell of spleen and bone marrow of wild type mice, B cell lymphoma cells in mice,reactive lymphoid hyperplasia issue in human, B cell lymphoma tissue in human, paraffin embedded lymphoma tissue without tumor metastasis and paraffin embedded B cell lymphoma tissue respectively. Remove genomic DNA in order to purify RNA and synthesis cDNA by reverse transcription.Screen these T-UCRs by qRT-PCR in B cell lymphoma cells of mice and bone marrow and spleen B cells of the normal mice respectively to find the T-UCRs which expressed highest Detect these T-UCRs by qRT-PCR in B cell lymphoma and the paraffin embedded tissues of human respectively,then compared the expression level of T-UCRs with that in the lymph node tissues which reactive hyperplasia and that in the paraffin embedded lymph nodes from the cancer specimens without cancer metastasis.Investigate the correlation between T-UCRs and B cell lymphoma at last.Results:T-UCR microarray:The expression of uc.88、uc.88A、uc.95、uc.167、uc.142、uc.282A、uc.300A、uc.398、 uc.399A、uc.316A、uc.268、uc.133A in B cell lymphoma cells of mice were higher than others.qRT-PCR:1.The expression of uc.316A and uc.95in B220positive B cells in bone marrow and spleen tissues of the wild type mice were significantly higher than that in B cell lymphoma cells of mice. And the expression of uc.268is on the contrary.2.The expression of uc.316A and uc.95in the lymph node tissues which reactive hyperplasia is significantly higher than that in B cell lymphoma tissue of human, so is paraffin embedded lymphoma tissue without tumor metastasis than paraffin embedded of B cell lymphoma tissue. And the expression of uc.268is on the contrary.Conclusion:Uc.316A and uc.95in mice and human B cell lymphoma could play an important role as tumor suppressor genes.Uc.268could be as an oncogene.Maybe these T-UCRs can be used as a molecular marker for differential diagnosis of B cell lymphoma, and can be used for the molecular targeted therapy.
