The study found, hypoadiponectinemia and development process of T2DM in hyperinsulinemia and insulin resistance. So some scholars propose, adiponectin can also be used as an independent factor of future diabetes patients. In the development of pathogenesis of periodontitis and diabetes, inflammatory disorders and aggravate is key, so by adiponectin periodontal intervention on patients with diabetic periodontitis may improve the periodontal status and overall health status in patients with diabetic periodontitis, reduce the occurrence and development of diabetes and its complications. However, adiponectin level can reflect the changes of periodontitis and local inflammation reaction insulin resistance develops, periodontitis involved in the mechanism of insulin resistance is associated with fat cell factor, these problems have not been resolved, and human studies also is unable to complete the study. Therefore, we adopt the method of Kunming mice at the age of3weeks to establish diabetic mice animal model of periodontitis, affecting the change of periodontitis research on adiponectin inflammatory state, and whether this effect is associated with insulin, so as to reveal the relationship between periodontitis and T2DM lesions, and provide a theoretical basis for clinical prevention and treatment of periodontitis.Objective:To detect the expression of adiponectin in diabetes periodontitis gingival in Kunming mice and the effects on gingival pathology and prognosis, so as to provide theoretical basis for clinical diabetes patients with periodontitis.Methods:Sixty3week old Kunming mice were divided into three groups according to the different molding methods:the control group (15rats), the diabetes group (15rats), the diabetes and periodontitis group (15), respectively, by four (Alloxan) on alloxan induced diabetic mouse model was established by injection of (12mg/100g body weight induced), mice model of periodontitis periodontal partial ligation combined with bacteria developed. By testing rats were injected with four oxygen pyrimidine, different time, blood glucose and fasting insulin level and insulin resistance index (Fins), and the blank control group compared to confirm the model success. By direct observations of inflammatory changes of gingival tissue, bone loss severity periodontitis model was successfully built calculation histopathological detection and Micro CT scanning. In after10weeks, three experimental groups of mice by direct observation of gingival inflammatory changes of gingival tissue, histopathology examination and Micro CT scanning calculation of bone loss in judging the degree of periodontitis. Content respectively by expression and three mice in the experimental group ELS A, RT-PCR, Western lot detection of gingival adiponectin gene and protein.Results:1.In this experiment,20day after modeling, blood glucose, fasting insulin level in diabetic patients and mice (Fins) is always greater than the normal value and the index of insulin resistance, and the blank control group was statistically significant difference (P<0.01), said the successful model. By direct observations of inflammatory changes of gingival tissue, bone loss severity periodontitis model was successfully built calculation histopathological detection and Micro CT scanning.2. the expression of adiponectin gene and protein in healthy mice gingival level was the strongest, in type2diabetes mellitus, type2diabetes patients with periodontitis group mice expression in gingivals weakened; differences were compared between groups, there were statistical significance (P<0.05).3three mice of the experimental group of gingival by direct observation of the detection of inflammatory changes and pathological gingival tissue display type2diabetes patients with periodontitis group mice gingival inflammation, the most obvious part of type2diabetic mice group showed moderate gingival inflammation, health mice without inflammatory gingivals; Micro CT scanning calculation of bone loss in the loss of the most obvious periodontitis gingivals of mice bone with type2diabetes, type2diabetic mice of group, healthy gingivals of mice did not see the bone loss. Conclusions:1. Effectively establish a mouse periodontitis model by partial ligation combined with periodontal bacteria, through four alloxan (Alloxan) injection was effective to establish diabetic mouse model.2. Compared with the healthy mice, type2diabetes patients with periodontitis group mice gingival inflammation and bone loss, the most obvious degree of adiponectin gene and protein expression in the lowest.3. Compared with the healthy mice, diabetic mice gingival inflammation and bone loss significantly, lower expression of adiponectin gene and protein.4. Results showed that the expression of adiponectin reduced gingival of diabetes is one of the important factors of periodontitis and aggravating, and provide a theoretical basis for clinical prevention and treatment of periodontitis.
