Objective:To analysis Vasohibin-1and VEGF-A, MVD expression levels and the relationship between the expression of Vasohibin-1and VEGF-A/MVD and the clinical pathological features in colorectal cancer tissues, explore Vasohibin-1to induce tumor mechanism in colorectal cancer, in order to provide guidance for the clinical diagnosis and treatment.Methods:60colorectal cancer patients who were underwent operation in The Second Hospital of Tianjin Medical University from Jun2013to Nov2013were included in this study. The Tumor tissue and tumor adjacent normal tissue mRNA expressions of Vasohibin-1and VEGF-A were detected by Real-time PCR (Reverse Transcription-Polymerase Chain Reaction). The protein expression of Vasohibin-1, VEGF-A and MVD were detected by immunohistochemical staining. The relationship between the expression of Vasohibin-1, VEGF-A and MVD, and the relationship between the expression of Vasohibin-1and clinicopathological features were evaluated in coloretacal cancer.Results:1. Real-time quantitative PCR detection of Vasohibin-1mRNA expression in colorectal cancer tissues was3.86times compare with the adjacent normal tissues; mRNA expression levels of VEGF-A is5.82times as much as it in the adjacent normal tissues, There were both statistically significant.(P<0.05).2. Immunohistochemical detection of60cases colorectal cancer tissue specimens, Vasohibin-1positive expression rate was43.3%(26/60), significantly higher than in adjacent normal tissue expression of16.7%(10/60). Vasohibin-1expression between the two groups was statistically significant (P=0.009). VEGF-A expression was51.7%(31/60), significantly higher than the adjacent tissues of18.3%(11/60), the difference was statistically significant (P<0.05). Microvessel density in colorectal cancer MVD (39.67±16.80/mm2) was higher than the value of adjacent tissue MVD (17.85±6.43/mm2). Vasohibin-1was closely related to VEGF-A protein expression in colorectal cancer (r=0.378, P=0.003) and was linear correlation with microvessel density (r=0.628, P<0.001).3. In60colorectal cancer patients, VEGF-A expression was related with TNM staging, distant metastasis and degree of differentiation (P<0.05). Vasohibin-1positive expression were closely related with distant metastasis and TNM staging of colorectal cancer (P<0.05).Conclusion:1. Colorectal cancer tissues Vasohibin-1, VEGF-A gene mRNA expression compared to adjacent normal tissue cells is upregulated.2. Vasohibin-1, VEGF-protein expressions are significantly higher in colorectal cancer tissues than in adjacent normal tissues.3. Vasohibin-1protein expression is correlated with the expression of VEGF-A, linear correlation with microvessel density in colorectal cancer.4. VEGF-A positive expression is related with TNM stage, distant metastasis and tumor differentiation in colorectal cancer patients,and Vasohibin-1positive expression is correlated with TNM stage and distant metastasis.
