ObjectiveTo study the relationship between cognitive function and expression of TNF-a, L-6factor in rat brain tissue with glucose tolerance abnormalities. The paper further explores the impact mechanisms of glucose tolerance abnormalities on cognitive function.MethodsHealthy male Wistar rats weighing180-200g are selected and randomly divided into group A or B with30in each group. Group A is a set of impaired glucose regulation model with high-sugar high-fat diet. Blood glucose in rats is measured after12weeks. When the model is determined to be successful, TNF-α, IL-6level in rat tissues with impaired glucose regulation will be measured in3W,6W,9W,12W and15W respectively by immunohistochemistry and situ hybridization, and the cognitive situation of rats evaluated by the water maze method in5given time points respectively. Group B is the normal control group without special treatment, but inflammatory factors level and cognitive function is measured and evaluated respectively in the3W,6W,9W,12W and15W. We analyze data with SPSS17.0statistical software and show them by mean±standard deviation (x±S). As for the compare of two sets of data, the count data is tested by t-test, and the measurement data tested with the chi-square test. When P<0.05, the result will be considered statistically significant.ResultsCognitive function assessment:The control group has no cognitive impairment and the test result is normal. But cognitive function assessment of impaired glucose regulation model group is significantly lower.Immunohistochemical determination of TNF-a:TNF-a of impaired glucose regulation model group has more expression at each time point and compared with the control group, TNF-a expression of impaired glucose regulation model group is higher at each time point (P<0.05).Immunohistochemical determination of IL-6factor:The result is similar to TNF-a. IL-6of impaired glucose regulation model group has more expression at each time point and compared with the control group, IL-6expression of impaired glucose regulation model group is higher at each time point (P<0.05).ConclusionThe expression level of inflammatory cytokines in rat tissues with impaired glucose tolerance is higher compared with normal control group, and the occurrence time of glucose tolerance abnormalities is positively correlated with expression of inflammatory cytokines.Cognitive disorders and inflammatory factors is significantly correlated, which further illustrate the relationship between incidence of cognitive impairment and inflammatory reaction.
