Monitoring Analysis Research Of Persistent Organochlorine Pesticides (OCPs) And Polychlorinated Biphenyls (PCBs) In Human Biological Samples

Persistent organochlorine pesticides (OCPs) and polychlorinated biphenyl (PCBs) have been produced and used in China in large quantities. There are different degrees of contamination of OCPs and PCBs in dietary and environmental media of China. OCPs and PCBs can accumulate in the body fat layer thus may cause serious health problems; therefore it is necessary to monitor the level of exposure to OCPs and PCBs in vivo.In the first part, a critical review is made on profiles of OCPs and PCBs production and use history in China, routes of human exposure to OCPs and PCBs, health effects may caused by OCPs and PCBs and detection methods of OCPs and PCBs in human biological samples. In the second (experimental) part, detection methods of40kinds of target OCPs and PCBs in human hair and breast milk were established, respectively. Then monitoring survey were conducted in75human hair samples of general population in Beijing and100blood samples. The results are as follows:1. Ultrafine grinding-accelerated solvent extraction-GC-ECD detection method of OCPs and PCBs in human hair samples was developed and validated. Limits of detection (LODs) of target OCPs and PCBs of established method were in the range of0.11to2.17ng·mL-1, limits of quantification (LOQs) were between0.36and7.25ng·mL-1; the average recoveries of spiked levels at50ng·g-1and100ng·g-1in human hair were91.2%and95.0%, respectively; RSD values were less than9.0%; inter-day and intra-day precision RSD values were less than8.0%. Besides, GC-ECD detection method of OCPs and PCBs in human breast milk samples based on ethyl acetate-cyclohexane (3:1, v/v) solvent extraction-PSA dispersive solid phase extraction cleanup was developed and validated. Average recoveries of spiked levels at20ng·g-1、40ng·g-1and100ng·g-1of all target compounds in human milk were81.4%,84.5%,98.2%, respectively; RSD values were less than5.0%, RSD values of intra-day precision were less than10.6%. Limits of detection (LODs) of target OCPs and PCBs of established method was in the range of0.lto2.3ng·g-1, limits of quantification (LOQs) was between0.2and7.7ng·g-1. To ensure the accuracy of the methods, matrix-calibrated solution was used to correct matrix effects error. HP-5and DB1701double-column were used to confirm the positive results. These two methods can be used for dynamic monitoring of OCPs and PCBs accumulate levels of human hair and breast milk. 2. Detection rates of target OCPs and PCBs in human hair samples (n=75) ranged from1.3to72.0%, with an average content of0.1to16.2ng·g-1. No significant difference was found between men and women of natural logarithmic transformation of the data detected of total HCHs, total DDTs, total chlordane and its derivatives, total OCPs, total indicative PCBs, total non-indicative PCBs, total PCBs, total PCBs and OCPs. And no significant correlation between OCPs and PCBs concentration in human hair with age was found.3. PCB209was not detected in blood samples (n=100). Detection rates of other target OCPs and PCBs ranged from1.0to92.0%, the average concentration of total PCBs was1.59ng·mL-1and total OCPs was9.28ng·mL-1. There are significant differences of exposure levels of tecnazene, y-HCH, quintozene, pentachloroaniline, heptachlor, dieldrin and PCB138between cancer and endocrine disease population and general population. Overall, levels of total composite exposure of40kinds of harmful substances in samples were low.
