Objectives:1.To investigate the influence of Gifitinib with5-Fu on cell proliferation and apoptosis on over-expressed HER-1gene in the human gastric cancer cells (SGC-7901) and to evaluate its antitumor genetics mechanism.2.To exolore the effects of the various processing methods (Gifitinib with5-Fu on over-expressed HER-1gene in the human gastric cancer cells) and the optimal scheme may be, so as to provide cytology basis for the clinical treatment.Methods:1. Gastric cancer cell lines SGC7901of her-1gene high expression was the research object.2. Detecting the inhibition rate of Gifitinib、5-fluorouracil respectively on human gastric cancer cells by MTT experiment, and then detecting the inhibition rate of the two drugs in different combination on gastric cancer cells by MTT experiment.3. Flow cytometric was used to determine apoptosis rate and the distribution of the cell cycle of Gifitinib and5-fluorouraci respectively on human gastric cancer cells or the two drugs in different combination on gastric cancer cell SGC-7901.4. RT-PCR method was used to detect the influence of Gifitinib、5-fluorouracil respectively or the two drugs in different combination to target protease mRNA on gastric cancer cells.Results:1.MTT showed:Gastric cancer SGC-7901cells was inhibited by Gifitinib or5-fluorouracil, with increasing dose of the drug, the inhibition rate of gastric cancer cells rised. The effect of chemotherapy drugs’cytotoxicity on over-expressed HER-1gastric cancer cells was augmented by Gifitinib. The best scheme was Gifitinib combined with5-fluorouracil and then sequential used targeted molecular medicine alone.2.Flow cytometric test indicated that compared with Gifitinib or5-fluorouracil group, the proportion of S period and G2/M period of the two drugs in different combination group was significantly reduced. a significantly different of apoptosis rate was found between the combination group and Gifitinib or5-FU alone.3. RT-PCR results showed that the combination group TSmRNA expression was significantly reduced compared with5-FU group Statistically significant (P<0.05). The inhibition of5-FU on TSmRNA can be enhanced by Gifitinib.Conclusion:Gifitinib in combination with5-fluorouracil in Her-1high expression human gastric cancer cell, inhibition increase apparently and results different between different scheme, the fourth scheme is the best.
