Objective:To investigate the dynamic expression of transcription factor Klf-4and its effect on acute inflammatory after the spine cord injuryMethods:Sixty SD rats were assigned into five groups at random.The5groups were sham-operation group (SHAM group, n=3), modeling control group,(MC group, T10injury,n-21),negative control group (NS group, T10injury+normal saline,5mg/kg,i.v., n=12), Honokiol group (HNK group, T10injury+HNK,5mg/kg,i.v.,n=12) and Methylprednisolone group (MP group, T10injury+.MP,5mg/kg,i.v.,n=12). The tenth thoracic vertebra (T10) was beaten and injuried by improved Allen’s Method. After modeling successful, MC group pick3rats at1h、2h、4h、6h、12h、24h、72h time point randomly to collect tissue from injured spine cord segment and test the cord samples with RT-PCR to measure the level of Klf-4. The other groups picked3samples at random to draw blood sample from aortaventralis and exam proinflammatory factors mRNA expression of Klf-4、NF-KB、p65、IL-6by RT-PCR and the content of chemokine-1(CXCL1) in blood by ELISA at6h、12h、241、72hResults:1.Klf-4mRNA expression increased from lh time point and went higher than SHAM group significantly at2h (P<0.01).then reached the peak at12h and decreased at24h,but still higher than SHAM group (P=0.02).2.Compared with the NS group, Klf-4level of HNK group was dropped remarkably (P<0.01).72h expression level decreased to the minimum in observable time window and lower the SHAM group, but not statistically significant (P=0.057).MP group showed Klf-4expression as much as NS group at6h (P=0.89) and12h (P=0.79) and indicated methylprednisolone didn’t inhibit Klf-4effective.3. NF-KB、p65、IL-6mRNA level in intervention groups showed an earlier raised and later decreased state in observable time window and had their peaks at12h as Klf-4. Each proinflammatory factors expression level of HNK group was statistically significantly lower than NS group at every time point (P<0.05).4. CXCL1concentration level in intervention groups also showed an earlier raised and later decreased state in observable time window and had their peaks at24h. HNK group was statistically significantly lower than NS group at every time point except6h (P=0.06). MP group expression level was between HNK group and NS groupConclusion:Klf-4gene and the other proinflammatory factors genes all appeared a dynamic expression with the same trend after spine cord injury. Honokiol can down-regulate Klf-4level which can be considered as its mechanism to decrease the other proinflammatory factors’expression. Klf-4might be the role as a gene and drug treatment target in acute spine cord injury.
