Abstract/Summary:
Lung cancer is one of the most common malignant tumors. Small cell lung cancer(SCLC) accounts for about15%to20%of primary lung cancer,。It was characterized by the lowest differentiation, highest malignancy, rapidly grow and early metastasis. When SCLC had been diagnosed lots of cases had transferred and lost the chance of operation. At present the main method of treatment SCLC is chemotherapy or radiotherapy or with combined treatment. Multidrug resistant (MDR) is the main reason for the treatment failure the of small cell lung cancer. How to detect drug resistance retreatment in SCLC at early stage and changed it. If by this method patient could be avoid overtreatment and then reduce the burden of the patients. Lots of studies suggest that the phenomenon of multidrug resistance is closely related with MDR1over expression. However the ideal target for reversal of multidrug resistance MDR1is still under research.Purpose:To explore the expression of PXR and MDRl in small cell lung cancer tissue and in peripheral blood then study their relationship and prognosis.Methods:①the expression of PXR and MDR1in40cases small cell lung cancer tissue with immunohistochemical method.②the mononuclear cells of peripheral blood(PBMCs) in31cases small cell lung cancer patients and20cases normal controls separated by Lymphocyte separation medium, the expression of PXR and MDR1of mononuclear cells with immunohistochemical method, Reverse-transc ription polymerase chain reaction(RT-PCR), realtime fluores-cence quantitative PCR was performed to detect(RTFQ PCR)Result:③The positive rates of PXR and MDR1in SCLC tissue were respectively35%17,5%. The expression of PXR and MDR1had no significant difference in the clinicopathological variable including age, gender, clinical stages (P>0.05).②The expression of MDR1was related with PXR(r=,0.49) inSCLC tissue③The expression of PXR, MDR1of small cell lung cancer patients was not statistically significant compared with normal controls in peripheral blood of SCLC(P>0.05)④The expression of PXR and MDR1were positively correlated before and after chemotherapy (r=0.562)Conclusion:①The expression of PXR and MDR1is different of in small cell lung cancer textural and peripheral blood.The expression of PXR and MDR1made it important in human small cell lung cancer multidrug resistance. PXR was more sensitive than MDR1in judging small cell lung cancer patients’chemotherapy response and prognosis.②PXR and MDR1expression in PBMCs may reflect the patient’s drug resistance phenotype and provide an easier method for MDR judgement in early stage of small cell lung cancer.
