Objective:To explore the pathogenesis and mechanism of preeclampsia.Methods:The T-bet and GATA3mRNA in PBMCs were detected by SYBR Green I real-timeRT-PCR. Results:The expression of GATA3mRNA in normal pregnancy group, mild preeclampsia group and severe preeclampsia group decreased with the aggravation of disease, there was significant difference between severe preeclampsia group and normal pregnancy group (P<0.05), but there was no significant difference between mild preeclampsia group and normal pregnancy group, severe pregnancy group (P>0.05); the expression of T-bet mRNA increased with the aggravation of disease, there was significant difference between severe preeclampsia group and normal pregnancy group, mild preeclampsia group (P<0.05); the expression of T-bet mRNA in mild preeclampsia group was higher than that in normal pregnancy group, but there was no significant difference (P>0.05);the ratio of T-bet mRNA/GATA3mRNA increased with the aggravation of disease, and there was significant difference between severe preeclampsia group and normal pregnancy group, but there was no significant difference between mild preeclampsia group and normal pregnancy group, severe pregnancy group (P>0.05)Conclusion:The high expression of T-bet mRNA which plays a role immunekilling and the normal expression of GATA3mRNA which plays a role immune protection or immune nutrition in PBMCs of patients with preeclampsia indicate that the high expression of T-bet mRNA in PBMCs is related to pathogenesis of preeclampsia and the effect of GATA3mRNA on occurrence and development of preeclampsia is not the same obvious as the T-bet mRNA, the ratio of T-bet mRNA/GATA3mRNA increased with the aggravation of disease, which indicated that Thl/Th2transforming growth factor imbalance may be one of the causes of preeclampsia.
