Objective:Establish the tumorous animal model with mice CT26 Colon Cancer Cells and induce multidrug resistance(MDR) by chemotherapy.Observe the status of tumorous growth and get the messages of the biological characters.Research the correlation of tumorous cells and peripheral blood mononuclear cells(PBMCs) expressive MDR1 before and after chemotherapyMethods:CT-26 colon cancer cells were injected subcutaneously into the right oxters of the mice.Fluorouracil(5-FU) and doxorubicin(ADM) were combined chemotherapy.Established mice tumor multidrug resistance model and falled into control group,49 day,70 day and 91 day groups with chemotherapy time.The statuses of tumouous growth were measured by tapy-B ultrasonic and sliding caliper.Drown growth curves and calculated doubling generation times,chemotherapy drugs sensitivity and drug fast index numbers of the tumor cells before and after chemotherapy by thiazol-z-yl-2,5-diphenyl tretrazolium bromide(MTT) method.Levels of MDR1b mRNA of the tumor cells and peripheral blood mononuclear cells were measured by reverse transcription polymerase chain reaction (RT-PCR).Eliminate function of P-glycoprotein was measured by flow cytometry(FCM).Results:Type-B ultrasonic showed the gross tumors volume were (0.63±0.11) cm~3 in the 27 day of the post inoculations from the control group,100%formed tumors.We measured nothing from the experiment groups in the 27 day.and in the 49 day the gross tumors volume were (1.51±0.50) cm~3 by sliding caliper,then the tumors developed fastness. Doubling generation times of the persisters and the parent cells had no statistics difference.Drug resistance indexes were 29.3,26.4,10.9,7.0,5.8,3.0,2.8 respective to ACT,ADM,VCR,HCPT,5-Fu,DAM,VP16 after chemotherapy and the drug sensitivities were lower.The levels of MDRlb mRNA increased after chemotherapy and P-glycoprotein excretive ability was higher than no chemotherapy group. The tumor and the PBMCs,MDR1b mRNA expression increased after chemotherapy and had surely correlation(coefficient correlation is 0.903) and P-glycoprotein excretive ability was higher than no chemotherapy group. Conclusion:The experiment of multidrug resistant animal model has established successfully,provided animal model for our study and research in the acquired multidrug resistance(AMR),and established groundwork for the research in elevation the chemotherapy effect.The leaves of MDR1b mRNA of the tumor and the PBMCs have correlation.The expression MDR1 is supposed to be a new kinetic index as evaluation the clinical chemotherapy of multidrug resistance and the prognosis of carcinomas.
