Objective:Using immunohistochemistry and PCR to detect IMP3, IGF-Ⅱ expression in cervical tissue.The study analyses the relationship between the expreesion level of IMP3and IGF-Ⅱ and clinical pathological features. This article explores the role and relevance of IMP3and IGF-Ⅱ in cervical cancer development,invasion and metastasis.Methods:1.Use immunohistochemical SP method to detect IMP3and IGF-II protein expression among125cases of cervical tissue (cases of cervicitis, CIN Ⅰ, CIN Ⅱ-Ⅲ and cervical cancer are respectively20,21,22and62).Then explored the relationship between the expression change and the clinicopathological paramenters.2.Use of real-time fluorescent quantitative PCR technique to detect IMP3and IGF-Ⅱ mRNA expression among65cases of fresh specimens of cervical tissue (cases of cervicitis, CIN Ⅰ, CIN Ⅱ-Ⅲ and cervical cancer are respectively6,8,11and40). Then explored the relationship between the expression change and the clinicopathological paramenters.3.Use SPSS19.0statistical software for statistical analysis on the experimental data, the inspection level of α=0.05.Results:1.The expression of IMP3in in cervical tissue(1)In cervicitis, CIN Ⅰ, CINⅡ-Ⅲ to cervical cancer, the IMP3protein positive rate are respectively10.0%、19.0%、59.1%'87.1%.The IMP3protein positive expression rate was significantly different in the four groups(P<0.05).From cervicitis, CIN Ⅰ, CIN Ⅱ-Ⅲ to cervical cancer, the IMP3increased gradually. Comparison between the four groups of two, CIN Ⅱ-Ⅲ group and cervicitis, cervical cancer group and cervicitis,cervical cancer group and CIN Ⅰ group, CIN Ⅱ-Ⅲ group and CIN Ⅰ, cervical cancer group and CIN Ⅱ-Ⅲ difference was statistically significant (P<0.0083); CIN Ⅰ group and cervicitis group comparison difference has no statistical significance (P>0.0083).(2)The expression of IMP3protein was not associated with patients’ ages, clinical stages, presence of high risk HPV, pathological type, presence of lymph node metastasis, and the degree of histologic (P>0.05).But the expression of IMP3protein was significantly related with patients’clinical stages(P<0.05).(3) In cervicitis, CIN Ⅰ, CINⅡ-Ⅲ and cervical cancer, the IMP3mRNA relative expression values are respectively1.770±1.298、2.551±2.100、4.926±2.844、11.820±6.876.From nomal cervical, CIN Ⅰ, CIN Ⅱ-Ⅲ to cervical cancer, the IMP3mRNA increased gradually.The IMP3mRNA relative expression values was significantly different in the four groups(P=0.000<0.05). Comparison between the four groups of two, CIN Ⅱ-Ⅲ group and cervicitis, cervical cancer group and cervicitis, cervical cancer group and CIN Ⅰ group, CIN Ⅱ-Ⅲ group and CIN Ⅰ cervical cancer group and CIN Ⅱ-Ⅲ difference was statistically significant (P<0.05); CIN Ⅰ group and cervicitis group comparison difference has no statistical significance (P=0.605>0.05).(4) The expression of IMP3mRNA was not correlated with patients’ ages, pathological type (P>0.05). But the expression of IMP3mRNA was significantly related with FIGO stage, presence of high risk HPV infection, presence of lymph node metastasis and the degree of histologic differentiation (P<0.05).2.The expression of IGF-Ⅱ in cervical tissue(1)In cervicitis, CIN Ⅰ, CINⅡ-Ⅲ and cervical cancer, the IGF-Ⅱ protein positive rate are respectively10.0%、14.3%、54.5%、83.9%, The IGF-Ⅱ protein positive expression rate was significantly different in the four groups(P<0.05).From nomal cervical, CIN Ⅰ, CINⅡ-Ⅲ to cervical cancer, the IGF-Ⅱ increased gradually. Comparison between the four groups of two, CIN Ⅱ-Ⅲ group and cervicitis, cervical cancer group and cervicitis, cervical cancer group and CIN Ⅰ group, CIN Ⅱ-Ⅲ group and CIN Ⅰ, cervical cancer group and CIN Ⅱ-Ⅲ group difference was statistically significant (P<0.0083); CIN Ⅰ group and cervicitis comparison difference has no statistical significance (P>0.0083).(2)The expression of IGF-Ⅱ protein was not significantly related with patients’ages, presence of high risk HPV infection, pathological type, and the degree of histologic differentiation (P>0.05). But the expression of IGF-Ⅱ protein was significantly related with patients’ FIGO stage and presence of lymph node metastasis(P<0.05).(3) In cervicitis, CIN Ⅰ, CINⅡ-Ⅱ and cervical cancer, the IGF-Ⅱ mRNA relative expression values are respectively1.256±0.702、1.574±0.843、3.127±1.962、7.721±5.326.From nomal cervical, CIN Ⅰ, CIN Ⅱ-Ⅲ to cervical cancer,the IGF-Ⅱ mRNA increased gradually.The IGF-Ⅱ mRNA relative expression values was significantly different in the four groups(P=0.000<0.05). Comparison between the four groups of two, CIN Ⅱ-Ⅲ group and cervicitis, cervical cancer group and cervicitis, cervical cancer group and CIN Ⅰ group, CIN Ⅱ-Ⅲ group and CIN Ⅰ cervical cancer group and CIN Ⅱ-Ⅲ difference was statistically significant (P<0.05); CIN Ⅰ group and cervicitis group comparison difference has no statistical significance (P=0.439>0.05).(4) The expression of IGF-Ⅱ mRNA was not significantly related with patients’ages, pathological type (P>0.05). But the expression of IGF-Ⅱ mRNA was significantly related with patients’clinical stages, presence of high risk HPV, presence of lymph node metastasis and the degree of histologic differentiation (P<0.05).3.Correlation analysis of IMP3and IGF-Ⅱ expression in cervical cancerThe result showed that from cervicitis, cervical intraepithelial neoplasia to cervical cancer, IMP3and IGF-Ⅱ expression both showed a trend of increase gradually. Their protein expression showed significant positive correlation(R=0.485, P<0.05). Their mRNA expression both showed a trend of increase gradually. IMP3and IGF-Ⅱ mRNA expression showed significant positive correlation(R=0.523, P<0.05).Conclusions:1.IMP3increased gradually with the increase of cervical lesion degree of trend. The expression of IMP3was both significantly related with patients’clinical stages, presence of high risk HPV, presence of lymph node metastasis and the degree of histologic. In conclusion, IMP3may play important role in the occurrence and development of cervical cancer.2. IGF-Ⅱ increased gradually with the increase of cervical lesion degree of trend. The expression of IGF-Ⅱ was both significantly related with patients’ clinical stages, presence of high risk HPV, presence of lymph node metastasis and the degree of histologic. In conclusion, IMP3may play important role in the occurrence and development of cervical cancer.3. The IMP3and IGF-Ⅱ has positive relevance in cervical cancer, both in the process of the development of cervical cancer may exist certain synergistic effect.
