ObjectivesThis project aims to explore the role of angiotensin Ⅱ and Interleukin-6in vascular injuryinduced by venous indwelling needle by measuring the contents of Ang Ⅱa nd IL-6in blood and vasculartissue through animal experiments as clinical application of intravenous catheter infusion simulation,.whichprovides a new basis for the clinical application of intravenous catheter and prevention of complications.MethodsTotally40New Zealand rabbits were divided into four groups randomly with10rabbits eachaccording to inclusion and exclusion criteria. one group is treated as the control group without anymeasures, and the venous blood will be extracted after one week’s conventional breeding and the middle ofvenous vascular tissue on the ears outer edge shall be extracted by anesthesia. Two edged-veins in both earsof other three groups were set intravenous indwelling needle as experimental group, normal saline20mlwas infused via the catheter every day. Used heparin saline sealed tube after injection. To extract plasma bycentrifuged after indwelling3days,5days and7days. and to store frozen specimens waiting for beingtested. Taking the rectangle organization with3cm pitch the puncture and each0.1cm wide of the centerline of both sides on puncture vascular after anesthetized. Drying with absorbent after the ice salineflush,weighing and taking150mg plus2ml cold saline to make liquid by homogenizer machine and storingthe frozen specimens to waiting for being tested. The contents of AngⅡ a nd IL-6in blood and vasculartissue were measured by radioimmunoassay instructions to observe the changes of its contents.Results1.The contents of Ang Ⅱ in plasma:5days’ group and7days’ were significantly higher than thatof the control group (P<0.01); the three groups are compared to show differences(P<0.01,P<0.05), andthe contents of Ang Ⅱ in turn increased with the the indwelling time extension.2.The contents of Ang Ⅱ in vascular tissue: there are significant differences in the wholeexperimental groups and control group(P<0.01); compared with the three experimental groups,5days’and7days’ groups are higher than3days’ group(P<0.05). 3.The contents of IL-6in serum:7days’ group were significantly higher than that of the controlgroup,3and5days’ groups(P<0.01).4.The contents of IL-6in vascular tissue:5days group and7days were significantly higher thanthat of the control group(P<0.01); and there are significant differences in the three experimental groupsthrough comparison(P<0.01).Conclusions1.The contents of Ang Ⅱa nd IL-6will be higher than that without puncture Intravenous catheter.2.The changes in Ang Ⅱa nd IL-6levels are related with the time by indwelling intravenouscatheter..3.The contents of Ang Ⅱa nd IL-6will be increased by set intravenous indwelling needle, itcauses vasoconstriction and spasm to injure vascular..
