Eucommiae Cortex is the dry barks of Eucommia ulmoides Oliv, and Eucommiae Folium is the dry leaf of Eucommia ulmoides Oliv. It is specially precious Chinese herbal medicines, which is distributed in Guizhou, Shanxi, Sichuan and other places. Both of them have been recorded in the first part of Chinese Pharmacopoeia2010version. Modern research has found that Eucommia barks and leaf have similar chemical composition and efficacy, with the efficacy of nourishing liver and kidney, strong bones and muscles, as well as the effect of the fetus.The results of this study are as follows:1. Comparing of the active ingredients’s content in three different extracting methods from Eucommiae Folium, it was determined by RP-HPLC. Chromatographic column:Thermo Hypersil Gold C18(250mm×4.6mm,5μm); the mobile phase was methanol-water-acetic acid(15:85:1); flow rate of1.0ml/min; and the detection wavelength was237nm. Results:When using the flash extraction, the content of chlorogenic acid was highest in Eucommiae Folium; the Geniposidic acid’s was highest by refluxing method. The content of Chlorogenic acid was1.63%,1.68%,1.76%by Heated to reflux, ultrasonic, flash extraction, and the content of Geniposidic acid was0.58%,0.59%,0.60%.2. The best extraction process of Eucommiae Folium extraction has been determined by using the flash extraction. Through the single factor experiment, using chlorogenic acid, geniposidic acid, as evaluation indicators, the70%ethanol was determined to the extract concentration of Eucommiae Folium. Through four factors three levels orthogonal experiment including extraction frequency, extraction time and solid-liquid ratio, concentration of ethanol extract, an optimum condition was obtained:70%ethanol extracts3times with6min for each; the ratio of material to liquid is1:25. Filtrating, the filtration was combined and concentrated in a relative density of1.05to1.10of clear paste, spray drying, the finished extraction was prepared.3. The quality standard of Eucommiae Folium extraction has been formulated. The means of using the chlorogenic acid as the reference substance of TLC identification has been established, and the protjects of moisture, heavy metal have been inspected. The HPLC fingerprint method for analyzing Eucommia Folium extraction was setted up. The optimum HPLC condition was elected on column of Thermo Hypersil Gold C18(250mm×4.6mm,5μm), and mobile phase of methanol and0.05%phosphoric acid in gradient elution, and temperature of25℃, flow rate of1.Oml/min, detector of UV with wavelength of254nm, the detective time was75min. Under the optimum HPLC condition, evaluating the fingerprints of Eucommia Folium extraction to generate control fingerprint in the different date, identified19common characteristics of the peak, and the content determination of the active ingredients such as geniposidic acid, chlorogenic acid were determinated.This article is researched about optimization of the extraction method of the active ingredients from Eucommiae Folium by Flash Extractor and prepared Eucommiae Folium extraction and quality standards. The results showed that:technology is reasonable and quality control method is advanced, extraction quality is stability, which provides experimental basis for the further research of the preparation.
