Objective: To investigate the effect of total flavone of Litchi ChinensisSonn (TFL) on the expression of TLR2、TLR4in hepatic fibrosis in rats, andto explore anti-fibrotic mechanism of TFL. Methods: Eighty males SD ratswere randomly divided into four groups:sham-operated group(SO), bile ductligation group(BDL), BDL with low-dose TFL treatmentgroup(100mg/kg/d, LDTFL),BDL with high-dose TFL treatmentgroup(200mg/kg/d,HDTFL).Exceptional SO group, the liver fibrosis whichwas induced by common bile duct ligation in rat was used as the animalmodel for this study,From the second day of operation, SO、BDL groupswere given physiological saline lavage, LDTFL, HDTFL groups were givenTFL lavage, all1times/day,4weeks.4weeks after operation all rats weresacrificed and the samples of livers were remained.Histopathologicalchanges in the livers were studied by Hematoxylin and Eosin (H&E)staining and Masson Trichrome (MT) examination. The expressions of TLR2and TLR4in liver tissue of above four groups were detected by IHCand WB.Results: The collagen area(μm2) detected by MT in SO group,BDLgroup,LDTFL group and HDTFL group were377.4±19.8,1172.5±108.4,1092.7±105.2,772.9±168.9,the area in SO groupwas significantly lower than those in BDL group (P<0.01),the area inHDTFL group was significantly lower than those in BDL group (P<0.01),thearea in LDTFL group and in BDL group have no different. Theexpressions of positive cells on TLR2and TLR4detected by IHC in eachgroup were:0.6±0.5、4.0±0.9、3.8±0.9、1.8±0.7and0.4±0.5、3.3±0.6、3.1±0.5、1.4±0.5,significantly lower in HDTFL group, in comparison withBDL group(P<0.01).The protein expressions of TLR2and TLR4detectedby WB in each group were:0.4±0.0、0.7±0.1、0.7±0.0、0.5±0.0and0.3±0.0、0.6±0.1、0.6±0.0、0.4±0.0,the protein expressions in HDTFL group weresignificantly lower than those in BDL group (P<0.01),the proteinexpressions in LDTFL groupwere between the BDL group and HDTFL group, however,therewas no significant difference compared with BDL group(P>0.05).Conclusion: The increase of TLR2and TLR4protein expressionmay have connected with the happening and development of liverfibrosis by CBDL in rat.TFL (200mg/Kg/d) can inhibit the expressionof TL2and TLR4in hepatic fibrosis in rats,and improve liver tissue fibrosis degree in CBDL rat.
