Objective:We will extracts active ingredients rfom Clausenalansium leaves, and establish asthmatic rat model,and measurethe concentration of cytokine IFN-y and IL-4which secreting byThl an Th2,study the effect of different dose of extracts rfom Clausena lansium leaves on controlling airway inflammation ofasthma rats and regulating the balance of Thl/Th2cytokinesrfom level of the whole animal expeirment,and thencomprehend the mechanism of effect on extracts from Clausenalansium leaves in asthmatic rat.MethodsrUsing ethanol relfuxing extraction to get the effectivecomponents from Clausena lansium leaves,and Using OVA toSensitize and challenge to established asthmatic rat model,byiflling the stomach with different concentration of Clausenalansium leaves, to observe the histopathology change of ratpulmonary tissues by hematoxylin and eosin(HE),to count thenumber of EOS,to measure the content of NO in rat lung tissuewith nitrate reductive enzymatic,and measure the concentrationof cytokine IFN-y> IL-4in serum of asthmatic rat by ELISA,andto analyze the expression of IFN-y> IL-4mRNA in asthmatic ratlung tissue was analyzed by RT-PCR.Results:(1)HE in lung tissue: asthma group showed there arelots of inflammatory cells infiltration round the bronchi,pulmonary alveoli and blood vessels.Mucous membraneswelling and hyperemiatenosis,wirnkle increased, lumens stenosisThere were no significantly inflammatory decreased changes inlow dose of extracts rfom Clausena lansium leaves group.But above-mentioned changes in middle and high dose of extractsrfom Clausena lansium leaves groups showed significantlydecreased changes. High dose of extracts rfom Clausenalansium leaves group inflammatory cells number is higher thandexamethasone group.However, we observed there were nosignificantly inflammation and normal lung tissue in controlgroup.(2)The count of EOS of blood: The count of EOS ofblood in asthma group were significantly higher than controlgroup (P<0.01),and different dose of extracts from Clausenalansium leaves groups were lower than athma group(P<0.05),but higher than control group (P<0.05).The count of EOS ofblood in high dose of extracts from Clausena lansium leavesgroup were lower than middle dose of extracts from Clausenalansium leaves group (P<0.05).The count of EOS of blood inmiddle dose of extracts rfom Clausena lansium leaves groupwere lower than low dose of extracts rfom Clausena lansiumleaves group (P<0.05).High dose of extracts from Clausenalansium leaves group EOS number is higher thandexamethasone group(P<0.05).(3)the content of NO in rat lungtissue with nitrate reductive enzymatic:The content of NO in ratlung tissue in asthma group were significantly higher thancontrol group(P<0.01),different dose of extracts from Clausena lansium leaves groups were lower than asthma group (P<0.05)but higher than control group (P<0.05).The content of NO in ratlung tissue in low dose of extracts from Clausena lansium leavesgroup were higher than middle dose of extracts from Clausenalansium leaves group (P<0.05).The content of NO in rat lungtissue in middle dose of extracts from Clausena lansium leavesgroup were higher than high dose of extracts rfom Clausenalansium leaves group (P<0.05).The content of NO High dose ofextracts rfom Clausena lansium leaves group is higher thandexamethasone group (P<0.05);(4)Measuirng the concentrationof cytokine IFN-丫、IL-4of serum and lung tissue by ELISA andRT-PCR: The expression of IL-4in serum and lung tissue inathmatic rats group detected significantly higher than controlgroup (P<0.01),and the expression in low dose of extracts rfomClausena lansium leaves group detected higher than the middledose of extracts rfom Clausena lansium leaves group (P<0.05);;he expression of IL-4in middle dose of extracts rfom Clausenalansium leaves group detected higher than the high dose ofextracts from Clausena lansium leaves group(P<0.05);theexpression of IL-4in high dose of extracts from Clausenalansium leaves group detected higher than dexamethasone group(P<0.05);The expression of IL-4from different dose of extracts rfom Clausena lansium leaves groups are higher than controlgroup (P<0.05);The expression of IFN-y in serum and lungtissue in athmatic rats group detected significantly lower thancontrol group(P<0.01), and the expression of IFN-y in high doseof extracts from Clausena lansium leaves group detectedhigher than the middle dose of extracts from Clausena lansiumleaves group(P<0.05); and the expression of IFN-y in middledose of extracts from Clausena lansium leaves group detectedhigher than the low dose of extracts rfom Clausena lansiumleaves group(P<0.05); the expression of IFN-y in high dose ofextracts from Clausena lansium leaves group detected lowerthan dexamethasone group (P<0.05);The expression of IFN-yfrom different dose of extracts from Clausena lansium leavesgroups are lower than control group(P<0.05).Conclusions:(1)Extracts from Clausena lansium leaves can inhibit theairway inflammation of asthma rats,the high concentrations ofClausena lansium leaves is more effective than the middleconcentration;the middle concentration of Clausena lansiumleaves is more effective than the low concentration.(2)Differentdose of extracts rfom Clausena lansium leavesinhibit the airway inflammation of asthma rats through regulating the balance of Thl/Th2cytokines.
