| Objective:Investigate the therapeutic effect of Rb1components in ginseng extract on hypertrophic scar through the hypertrophic scar on the rabbits’ears. Methods:Select15rabbits (male or female) in the middle of each rabbits’ear (2～3), about1cm circular defects (there are5on each rabbit). Let it have a natural healing process. Treat each scar with different methods after26days later. Grouping them:blank control-group a, physiological saline-group b, Rbl-group c (0.2mg/ml)、d (0.4mg/ml)、e(1.0mg/ml). The dose is about0.3ml, injection drug1times every3days, a total of3times. Second and drug injection after the end of the week, forth weeks and sixth weeks. These scars put into paraformaldehyde. After one month, the scar tissues become paraffin sections, such as HE staining, VG staining, microscope to measure and calculate the scar index (HI), collagen, immunohistochemistry to detect the expression of Caspase-3and in situ hybridization method to measure collagen I mRNA. The data were processed by SPSS13.0, the measurement data with " x±s" form, for t test and variance analysis, P≤0.05with significant difference..Results:With the increase of drug concentration, HI and collagen concentration decreased. But there are no statistical significance between c and d, P>0.05, D, e compared with C and d had statistical significance, P<0.05.That means high concentration of drugs can reduce hyperplasia. The same situation also be found in the expression of Caspase-3immunohistochemical and in situ hybridization of Collagen I mRNA, high concentration effect of ginsenoside Rbl on Caspase-3increased, mRNA decreased the expression of collagen type.Conciusion:Ginseng extract Rbl can reduce the synthesis of collagen in the hyperplastic scar, promote fibroblasts apoptosis.At last, inhibit the development of hyperplastic scar effectively. |
