Objective:To explore the mechanism of hypertrophic scar from rabbit ear model, and observe the change of PDGFR-a,PDGFR-βand MMP-1 after use Kangfuxin solution, Silicone oil urea dexamethasone cream and Kangfuxin solution with Silicone oil urea dexamethasone cream. To provide a scientific basis for further development of effective drugs for treatment of hypertrophic scar.Methods:Forty-eight three-months-old New Zealand rabbit were randomly divided into six different groups:blank group, model group, positive control group, Kangfuxin solution group, Silicone oil urea dexamethasone cream group, Kangfuxin solution with Silicone oil urea dexamethasone cream group. After medication corresponding drugs for 14 days and 28 days, use HE staining to observe the hypertrophic scar general structure, hypertrophic scar index and the number of fibroblasts. Use Masson method to observe density of collagen fibers. Use Immunohistochemical method to observe the change of PDGFR-a, PDGFR-βand MMP-1.Results:Compared with model group, the expression of scar thickness in all treatment groups was down-regulated after medication corresponding drugs for 14 days and 28 days, the result was significant difference and the difference has statistical significance (P<0.01), kangfuxin solution with Silicone oil urea dexamethasone cream group has the most significant difference. Compared with model group, the expression of HI in all treatment groups was down-regulated after medication corresponding drugs for 14 days and 28 days, the difference has statistical significance(P< 0.05 or P<0.01), silicone oil urea dexamethasone cream group and kangfuxin solution with Silicone oil urea dexamethasone cream group have the most significant difference. Compared with model group, the expression of fibroblasts and density of collagen fibers in all treatment groups was down-regulated after medication corresponding drugs for 14 days and 28 days, the difference has statistical significance(P<0.05 or P<0.01). Compared with model group, the expression of PDGFR-a,PDGFR-βand MMP-1 in all treatment groups was down-regulated after medication corresponding drugs for 14 days and 28 days, the difference has statistical significance(P<0.05).Conclusions:All of the treatment groups can reduce the fibroblasts, density of collagen fibers, thickness of the hypertrophic scar of rabbit ears, and the kangfuxin solution with silicone oil urea dexamethasone cream group has the most obvious effect. Regulation of PDGFR-a, PDGFR-β and MMP-1 to reduce the excessive deposition of hyperplasia of fibroblasts and collagen, so as to achieve the prevention and treatment of hypertrophic scar.
