BackgroundGliomas are the most common malignant tumors of the central nervous system,accounting for40%‐50%of intracranial tumors.In recent years, although the terms of the skill level of neurosurgery, radiotherapy and chemotherapy drugs and other equipment have made considerable progress.However, as with invasive growth of glioma and normal brain tissue adhesion phase decomposition is unclear, for highly resistant to conventional chemotherapy, radiotherapy and other characteristics, the treatment effect is not ideal.Statistics show that only less than30%survival of glioma patients can be more than two years.Therefore, to study the pathogenesis of gliomas at the molecular level, explore new treatments and technologies to provide a new path for the clinical diagnosis and treatment of gliomas.Objective:Generally, the pathogenesis of gliomas is considered to be a multi‐factorial and multi‐step process.Signal transduction and activator of transcription3(signal transducer and activator of transcription3, STAT3) are cytoplasmic protein family, having a signal transduction, transcriptional regulation dual function.P16is a tumor suppressor gene, also known as MTS (multipletumorsuppressor1) gene, a new anti‐cancer gene was found by Kamb and other U.S. scientists in1994.It is involved in cell cycle regulation, a negative regulator of cell proliferation and division.The purpose of this experiment is to study the expression of STAT3and P16in gliomas, analysis their deference and correlation, In order to further explore the relationship between the two expression in human brain glioma, provide reference for clinical diagnosis and treatment.Materials and Methods:This experiment collected75patient’s tumor tissue and clinical data who were surgery at the First Affiliated Hospital of Zhengzhou University during December2012to November2013.According to the pathological test results,which grade Ⅰ17cases, Ⅱ20cases, Ⅲ grade18cases, grade Ⅳ20cases.According to WHO (2000) classification of tumors of the central nervous system, the grade Ⅰ, Ⅱ grade into low‐level group, grade Ⅲ, Ⅳ grade into the high‐level group.In addition the department collected10cases within the same period due to severe head trauma decompression surgical resection of brain tissue as a normal brain tissue (biopsy were confirmed by pathologists validation).Take SP immunohistochemical method to detect the expression of STAT3and P16intensity in gliomas, then χ2test and spearman correlation analysis of STAT3and P16in levels of expression in gliomas, the last and then spearman rank correlation analysis of the correlation between STAT3and P16further explore the links between the two.Results1. STAT3:The expression of STAT3was negative in all10cases of normal brain tissue.In glioma, as the pathological level increased, the expression of stat3gradually enhanced:Low level group was37.84%(14/37); High level group was 71.05%(27/38). The difference between the low‐level and high‐level group was significantly.2. P16: The expression of P16was positive in10cases of normal brain tissue. In glioma, with the increase of pathological level, the expression of P16gradually weakened: Low level group was78.4%(29/37); High level group was28.95%(11/38). The difference between the low‐level and high‐level group was significantly.3. The relationship between STAT3and P16: According to the spearman correlation test, as the pathological level increased, the expression of stat3gradually enhanced and the expression of P16gradually weakened. Their differences were statistically significant,(rSTAT3=0.452, p <0.05; rP16=‐0.524, p <0.05); The expression of Stat3protein and P16protein in human glioma showed a negative correlation.(r=‐0.937, p <0.05).Conclusions:STAT3signaling pathways and P16associated with the malignant degree of human brain glioma. as the pathological level increased, the expression of stat3gradually enhanced and the expression of P16gradually. Thus we can speculate that STAT3and P16play an important role in the development process in human brain glioma, combined detection of STAT3and P16expression levels can be used as an reference index for evaluation of the biological behavior and prognosis of glioma. We also can give the corresponding single or joint targeted therapy, to further promote the improvement of treatment and clinical effect.
