Aberrant transduction and regulation of cell signaling keep close with the occurrence and progression of tumor. Some signaling pathways are centered on a few of transcription factors, which are the final 'switches' that ultimately lead to cell malignant transformation. Some studies have shown that the signal transducer and activator of transcription 3(Stat3) signaling is constitutively activated in a great deal of tumors and plays an important role in oncogenesis and progression of tumors by abnormally regulating the cell survival and transformation. Stat3 singnaling has been a hot spot of research nowadays.Stat3 signaling pathway is activated after receiving extracellular signals such as epidermal growth factor, cytokines. Activated Stat3 (phosphorylated-Stat3) enters cellular nucleus and binds to specific DNA promoter to regulate target genes transcription which control fundamental physiologic processes such as cell proliferation, apoptosis and angiogenesis. In normal condition, activation of Stat3 signaling pathway is temporary and is controled accuratly. But with various kinds of tumorigenesis signal stimulation, Stat3 presents in the cell nucleus permanently and regulates target genes transcription continuously being involved in malignant transformation of tumor cells, called constitutively activation of Stat3 signaling pathway.
