Purpose: Through experimental study observed Tiaozhi Tongluo Capsule onthe rat ventricular remodeling after myocardial infarction and myocardialtissue morphology RAAS system PRA, Ang Ⅱ, ALD、ANP、BNP、TNF-a、IL-6and other indicators, analyzes the efficacy mechanisms may exist.Method:120Male Sprague Dawley rats were randomly divided intoSham-operation group (n=10) and AMI group (n=110). AMI was establishedby ligation of LAD and the sham-operation rats underwent the same produreexcept for the coronary artery. After24hours, the survival AMI rats weredivided randomly into Model group, ATO group, CAP group, TXL group,TZTL group. The medicine interfere groups were treated with medicine for4weeks, once a day, The dosage of rats (mg/kg/d)=The dosage of human(mg/kg/d)/60kg×6.3. While the Sham and the Model group were treated withplacebo (saline, i.p.). After4weeks’ treatment, The morphological changesobserved in rats with myocardial infarction area, and serum lipids weredetected (TC, TG, LDL, HDL), PRA, Ang Ⅱ, ALD, ANP, BNP, TNF-a, IL-6levels and other indicators of change.Results:1. Successful model of myocardial infarction, survival after the end of theModel group47rats, in which the Sham group10.2. After4weeks, Compared with Sham group, TC, TG, LDL-C in Modelgroup was increased, and HDL-C was reduced (P<0.05). Compared withModel group, TC, TG, LDL-C in ATO group, TXL group, TZTL group wasreduced, and HDL-C was increased (P<0.05).Compared with Model group, TC,TG, LDL-C in CAP group was reduced (P<0.05). The difference was notstatistically significant between TZTLgroup with positive control groups (P>0.05).3.After four weeks of drug intervention, TZTL group compared with theSham group, the lung wet to dry ratio, lung water content were significantlydifferent (P>0.05). TZTL group compared with the Sham group scar thickness,left ventricular internal diameter has changed significantly, with statisticalsignificance (P <0.05); compared with the control group was significantlythicker scar thickness, left ventricular internal diameter decreases significantlydifferent (P <0.05)。4. Sham group showed a very small amount of PRA, Ang Ⅱ, ALD, ANP,BNP, TNF-a, IL-6serum concentration signs, Model group PRA, Ang Ⅱ,ALD, ANP, BNP, TNF-a, IL-6serum concentrations higher than Sham groupwas significantly higher, the positive control group PRA, Ang Ⅱ, ALD, ANP,BNP, TNF-a, IL-6serum concentrations decreased compared with Modelgroup, with statistical significance (P <0.05). ATO group PRA, Ang Ⅱ, ALD,ANP, BNP, TNF-a, IL-6serum concentrations compared with Sham groupdifference was not significant (P>0.05), TZTL group than in the Model groupPRA, Ang Ⅱ, ALD, ANP, BNP, TNF-a, IL-6serum concentrations decreasedsignificantly (P <0.05), no significant difference (P and the positive controlgroup>0.05).Conclusion:1. An ideal model of AMI model can be established by ligation of LAD inSprague Dawley rats.2. Tiaozhitongluo Capsule can improve dyslipidemia, downregulate theserum levels TC, TG, LDL-C.3. Reduced survival in rats after myocardial infarction, left ventricularinternal diameter enlarged scar thinner, could be the next after MI stimulationof various factors, rat serum PRA, Ang Ⅱ, ALD, ANP, BNP, TNF-a, IL-6content increased, prompting the occurrence of ventricular remodeling,increased cardiac load and stimulate cardiac hypertrophy, exacerbation ofchronic cardiac load conditions, resulting in changes in cardiac morphologyand function. Capsule can improve lipid through the survival of ratsmyocardial infarction, left ventricular internal diameter narrowing, thickeningof myocardial scar thickness, after taking into account the rat myocardialinfarction PRA, Ang Ⅱ, ALD, ANP, BNP, TNF-a, IL-6inhibition may be generated.4. After lipid capsules meridians can improve heart function in ratmyocardial infarction, post-myocardial infarction in rats have lower serumPRA, the role of Ang Ⅱ, ALD, ANP, BNP, TNF-a, IL-6levels, effectivelyreducing large myocardial infarction rat mortality, ventricular remodeling afterits mechanism may be decreased serum PRA, Ang Ⅱ, ALD, ANP, BNP,TNF-a, IL-6levels of inhibition of myocardial infarction achieved.
