LV remodeling is defined by alteration in LV size, shape, and wall thickness after an AMI. People consider that the patho-foundation of chronic cardiac failure was ventricles reconstitution. Preventing and reversing the ventricular remodeling becomes the important turning point in clinical treatment. Collagen network remodeling is the key point in tissue fibrosis after AMI. Recently research have reported that TGF-β1,MMP-2 and MMP-9 correlate compactly with ventricular remodeling after AMI and the metabolism of collagen. There are empirical studies and clinical applications showed that the Yindan Xinnaotong Capsule can improve myocardial ischemia, increase coronary flows, and expand the constrictive coronary vessels, but there was not enough research on ventricular remodeling after acute myocardial infarction.This study was investigate the effects of TGF-β1,MMPs on collagen network remodeling and the effects of"Yindan Xinnaotong Capsule"on ventricular remodeling after acute myocardial infarction in rats.Methods: AMI was established by ligating of the left anterior descending coronary artery in WS rats. 24 hours after the procedure, the surviving rats were separated randomly into four groups: AMI group,high dose of"Yindan"group,low dose of"Yindan"group,captopril group, sham-operated rats were selected randomly as non-infarction control. The cardiac muscle samples were obtained four weeks after AMI to detect the expression of TGF-β1,MMP-2,9 mRNA in noninfarcted zone by reverse transcription polymerase chain reaction (RT-PCR), then the rats were sacrificed to determine the left ventricular weight and heart index,Myocardial cell and interstitial tissue were observed by HE stain and Masson stain., and stain myocardial tissue with HPIAS1000 type medical image analytical system to detect collagen volume fraction (CVF).Results: Compared with those of sham-operated group, the left ventricular weight and heart index, and the expression of TGF-β1,MMP-2,9 mRNA were all increased significantly(P<0.01) in AMI group after 4 weeks. Compared with AMI group those parameters were all decreased obviously in treatment group, the high dose of"Yindan"group decreased significantly, and there were no obvious differences between low dose of"Yindan"group and captopril group. Conclusions:"Yindan Xinnaotong Capsule"Could obviously Reducing the expression of TGF-β1,MMP-2,9 in myocardial tissue, by which reduces VR after AMI in rats.
