ObjectiveThis study aims:Taking ulcerative colitis rats as investigative objects, selecting the intestinal molecular biological immune and the volts poison theory of TCM as the pointcut, observed the influence that TCM clear away volts poison treatment under the guidance of the volts poison theory to UC rats’ intestinal mucosa and plasma plasma levels of MMP-1and TIMP-1, While collecting clinical cases of ulcerative colitis, for observing syndrome differentiation of TCM, examined the relationship between serum MMP-1, TIMP-1levels between the different TCM syndrome types. To discusse the influence between the volts poison theory to UC rats at the level of molecular biology and the relationship between different syndrome types of poison. Conducted clinical research with the prescriptions-disease as carrier, provided the objective evaluation to the relationship between the volts poison theory of TCM and ulcerative colitis, expounds the curative effect of TCM, and promoted the theory of TCM rigorous.MethodsTo take180-220g40SD rats. All of the rats randomly were divided into blank control group(n=10), Tiaochang Xiaoyan Tablets group(n=10), SASP treatment group(n=10), distilled water treatment group(n=10). In addition to the blank control group, the rest of rats use the composite method of dextran sodium sulfate(DSS) to build module7days. After7th day, Tiaochang Xiaoyan Tablets group fills medicine of mixed suspension10ml/kg, SASP treatment group fills SASP of physiological saline45mg/ml, distilled water treatment group fills distilled water lml/100g, all groups treatment of21days. After building module and treatment, we detecte the body weight of rats and stool shape, fecal occult blood test. We observe the intestinal mucosa and plasma levels of MMP-1, TIMP-1before and after treatment. The rats of Tiaochang Xiaoyan Tablets group after the treatment of21days were relieved symptoms, keep treatment7days, then observing the intestinal mucosa and plasma levels of MMP-1and TIMP-1. At the same time,we collect42clinical cases (with12cases in large intestine Damp-Heat syndrome group,12cases in mixed Cold and Heat syndrome group,12cases in Spleen Qi deficiency group and6normal people for normal control group). After then, the TCM syndrome was observed, we maked TCM syndrome differentiation, and detection in patients with serum levels of MMP-1and TIMP-1. We will input the collected data to the SPSS17.0packets of data, In the homogeneity of variance and the premise of normal distribution, the measurement data between two groups were compared with t test. The measurement data in multiple groups were compared with One-way analysis of variance and the comparison among groups were performed with SNK or LSD test. The count data were compared with by chi-square test (or exact probability test).Results1. Experimental animalsection(1) MMP-1, TIMP-1levels in rats after modelingThe MMP-1, TIMP-1levels of model rats(Tiaochang Xiaoyan Tablets group, SASP treatment group, distilled water treatment group) were significantly higher than the blank control group rats (P<0.05), the difference was statistically significant, but the MMP-1, TIMP-1levels in modules were not significantly different (P>0.05).(2) The changment of MMP-1and TIMP-1levels before and after treatment in within group ratsThe MMP-1, TIMP-1levels were significantly decreased after treatment in Tiaochang Xiaoyan Tablets group and SASP treatment group rats. The difference was statistically significant (P<0.05), the treatment is effective. The MMP-1, TIMP1levels of the blank control group rats were showed no significant difference after treatment (P>0.05). While the MMP-1levels in distilled water treatment group rats after treatment was not statistically significant difference (P>0.05), TIMP-1levels after treatment compared with the previous fall (P<0.05).(3) The MMP-1and TIMP-1levels difference comparison between groups before and after treatmentThe MMP-1, TIMP-1decreased in value in Tiaochang Xiaoyan Tablets group and SASP treatment group rats were no significant difference (P>0.05), these two groups were compared with distilled water treatment group:MMP-1decreased in value was statistically significance difference (P<0.05), TIMP-1decreased in value was not statistically significant difference (P>0.05).(4) The MMP-1, TIMP-1levels in continued treatment after UC rats getting remissionThe Tiaochang Xiaoyan Tablets group rats were in remission after21days of treatment, further treatment7days MMP-1, TIMP-1levels decreased compared with the treatment of21days, the difference was statistically significant (P<0.05).2. Clinical observation section(1) Baseline analysisBy the statistical tests, the sex and age of patients in each group were no statistical difference(P>0.05).(2) The MMP-1, TIMP-1levels in different TCM syndromesComparition between each group, we found that the MMP-1, TIMP-1level’s of mixed Cold and Heat syndrome group were lower than that in large intestine Damp-Heat syndrome group (P<0.05), but higher than the Spleen Qi deficiency group(P<0.05), the difference was statistically significant. The MMP-1, TIMP-1levels in these three groups were higher than normal control group, the difference was statistically significant (P<0.05).Conclusion(1) UC pathogenesis is related with MMP-1, TIMP-1levels.(2) The MMP-1, TIMP-1levels can be used as external indicator of objective evaluation in the volts poison theory. It’s effective in volts clea by the volts poison theory to UC rats, and efficacy like SASP. Efficacy was reflected in reducing the MMP-1, TIMP-1levels. In remission also have a certain effect.(3) There are some difference in the level of serum MMP-1, TIMP-1between the different TCM syndromes is different in clinical.It’s highest in large intestine Damp-Heat syndrome group, then the Cold and Heat syndrome group, followed by the Spleen Qi deficiency group. But these three groups were higher than normal control group.(4) MMP-1, TIMP-1can be used as an outward manifestation of volts poison in molecular level.To some extent, we realized the relationship objectification between volts poison theory and UC.
