Objective:Mast cells (mast cell) is widely distributed in the connective tissue of skin and visceral mucous membrane, has strong basophilic granular cells. Secretion of cytokines, involved in immune regulation, abundant expression of IgE receptor Fc, the release of allergic media. Mast cells and substance particle release, containing heparin, histamine,5-hydroxytryptamine, can cause anaphylaxis. The mast cells obtained from SD rats abdominal cavity, observe the fish up grass (Houttuynia cordata Thunb.) extract on Anti-DNP IgE induced mast cell degranulation, histamine release, intracellular calcium intake. To observe the effects of vascular permeability fish woke up grass extract on Anti-DNP induced by IgE in rat skin allergic reactions.Methods:Induction of rat skin allergic reaction by Anti-DNP IgE (PCA) experiment, using the Evans blue dye to observe the vascular permeability; degranulation of mast cell induced by using Anti-DNP IgE, collected from rat peritoneal mast cells, and divided into five groups, normal group, model group, low concentration group of Houttuynia cordata Thunb extract (30mg/ml); concentration of Houttuynia cordata extract in (60mg/ml); Houttuynia cordata Thunb extract of high concentration group (120mg/ml), by measuring the effect of Herba Houttuyniae on mast cell assay, in vitro experiments of Houttuynia cordata Thunb extract on Anti-DNP IgE induced mast cell degranulation, histamine release, effect of intracellular calcium intake.Results:The concentration of extractive of Houttuynla cordata thunb in30~120mg/mL had no significant effect on the absorbance of the mast cells. It suggested that extractive of extractive of Houttuynla cordata thunb had no obvious effects on the survival of mast cells. Mast cell degranulation was induced significantly by anti-DNP IgE. Mast cell degranulation was not inhibited by30mg/mL extractive of Houttuynla cordata thunb pretreatment, but mast cell degranulation was not obvious and significantly inhibited by the pretreatment of extractive of Houttuynla cordata thunb in60or120mg/mL. Histamine generation was very low in normal group, which was significantly increased in degranulation group compared with in normal group(P <0.05). Histamine generation was reduced by all of extractive of Houttuynla cordata thunb treatment group, which was statistically significant in high or middle concentration group (120,60mg/ml) compared with in degranulation group (P<0.05). Evans blue exudation was little in normal group, which was significantly increased in model group compared with in normal group (P<0.05). The exudation of dye was reduced by all of extractive of Houttuynla cordata thunb treatment group, which was statistically significant in high or middle dose group (300,150mg/kg) compared with the model group (P<0.05). Calcium intake was little in normal group, which was significantly increased in degranulation group compared with in normal group (P <0.05). Calcium intake was reduced by all of extractive of Houttuynla cordata thunb treatment group, which was statistically significant in high or middle concentration group (120,60mg/ml) compared with the degranulation group (P<0.05). Conclusions:1. Mast cells degranulation and histamine release was significantly inhibited with extractive of Houttuynla cordata thunb treatment in rats.2. plasma leakage mediated by mast cells was significantly inhibited with extractive of Houttuynla cordata thunb treatment in rats.3. Mast cells degranulation was significantly inhibited with extractive of Houttuynla cordata thunb treatment in rats through calcium influx decrease in mast cells...
