Chinese medicine quality standard is an important part of the national drug standards. The species of Chinese herbs are various and chemical composition is complicated. The existing quality standards still have some defects in the quality control and specificity of traditional Chinese medicine which can only be limited to qualitative identification of herbs and quantitative detection of a single component. The problems of adulteration are not strictly tested in the quality standards. In additional, any ingredient of traditional Chinese medicine products during measurement and control period can not represent its overall quality. This paper systematically conducted in-depth research to the medicine quantitative microscopic method which aims to apply a new way, namely quantitative microscopic method, as a supplement to chemical methods, to test the quality of the traditional Chinese medicine.Purpose:To establish a improved quantitative microscopic method and to test the content of Panax ginseng, American ginseng, Notoginseng cilster crystal, Resin canal, catheter, Cork cells in the Araliaceae Chinese medicine and the content of rhubarb, Notoginseng, Polygonum cilster crystal, catheter, Cork cells, fiber, Brown blocks, stone cell in the Polygonaceae medicine. Analyze the microscopic content of each medicine comprehensively to provide a reference for the quality of medicine identification. With saponin as an index component for Araliaceae and with anthraquinone for Polygonaceae, determine the total content of saponins or anthraquinone in various herbs respectively. Then conduct correlation analysis between the composition and microstructure features to examine whether there is a linear correlation existed.Methods: First, establish improved quantitative microscopic method. Then, Chloral hydrate, with the physical properties of the low melting point, can be used as the solvent of mixed samples. After the medicinal powder was added into melting chloral hydrate, cooling down immediately to get the solid mixed samples. Apply quantitative Morphometric methods in stereologies to measure the quantity of several microscopic characteristics.Analyze multiple microscopic characteristics indicators of all the herbs with linear regression method and demonstrate the method thoroughly to achieve the aim of determining the quality of traditional Chinese medicines. The microscopic characteristics of each medicinal traits size, measured by particle size measurement method in stereologies, could be adjutant basis for quality identification.Test the total content of saponin in Panax, ginseng, rhubarb, Polygonum cuspidatum with UV-visible spectrophotometry method and the content of anthraquinone in Rhubarb, Polygonum cuspidatum, Polygonum. Then examine the correlation between the microscopic characteristics of various herbs and their total chemical composition.Result:1.the amount of each medicine and microscopic character indicators measured by improved quantitative microscopic method showed a significant linear correlation, P was less than0.05. The amount of microscopic characters in Panax ginseng, American ginseng, Panax, Polygonum cuspidatum, rhubarb and Polygonum are differ by different sources.2.①Moderate correlation existed between the microscopic character content of crystal clusters and saponin in Panax ginseng, Panax quinquefolium L and Notoginseng;②Low correlation existed between the microscopic character content of Resin canal and saponin in Panax ginseng and Panax quinquefolium L, but high correlation existed between the microscopic character content of Resin canal and saponin in Notoginseng, P is lower than0.05which has statistical significance.③Low correlation existed between the microscopic character content of catheter and saponin in Panax ginseng and Notoginseng, moderate correlation existed between the microscopic character content of catheter and saponin in Panax quinquefolium L;④Low correlation existed between the microscopic character content of Cork cells and saponin in Panax ginseng and Notoginseng, and high correlation existed between the microscopic character content of Cork cells and saponin in Panax quinquefolium L.3.①High correlation existed between the microscopic character content of crystal clusters and anthraquinone in rhubarb, Polygonum cuspidatum, Polygonum. P is lower than0.05which has statistical significance;②Moderate correlation existed between the microscopic character content of catheter and anthraquinone in rhubarb and Polygonum, but high correlation existed between the microscopic character content of catheter and anthraquinone in Polygonum cuspidatum, P is lower than0.05which has statistical significance;③Low correlation existed between the microscopic character content of Cork cells and anthraquinone in rhubarb and Polygonum, but high correlation existed between the microscopic character content of Cork cells and anthraquinone in Polygonum cuspidatum. P is lower than0.05which has statistical significance④Low correlation existed between the microscopic character content of fiber and anthraquinone in rhubarb, Polygonum cuspidatum, Polygonum.⑤High correlation existed between the microscopic character content of Brown blocks and anthraquinone in Polygonum cuspidatum. P is lower than0.05which has statistical significance. Moderate correlation existed between the microscopic character content of Brown blocks and anthraquinone in Polygonum.⑥Moderate correlation existed between the microscopic character content of stone celland anthraquinone in Polygonum cuspidatum.Conclusion:The microscopic character content of several herbs and indicators measured through the improved quantitative microscopic method is stable and reliable which can comprehensively reflect the similarities and differences of various microscopic character amount. The measurement result of typical microscopic character in the various herbs of Araliaceae and Polygonaceae shows that the microscopic characters of calcium oxalate cilster crystal, Resin canal, stone cells and brown blocks are distinct. The cilster crystal of powder medicinal material remains an overall which is easily identified and can be used as indicators of the microscopic character to test the medicinal material contained calcium oxalate cilster crystal. The result measured is stable and reliable. affected by crushing, screening, etc, the cilster crystal of catheters, cork cells and fibers has various size and could be used as adjutant indicators for microscopic quantification of Chinese medicine materials. The amount of the microscopic character of one Chinese medicine material can be used as an adjutant basis for quality identification of that medicine material.The analysis of the correlation of microscopic characters and ingredients indicates that:some relevance existed between some microscopic character of Chinese herbal medicines and their chemical ingredients. the microscopic character contents can be further controlled under the requirements of the chemical composition to achieve the objective of distinguishing the quality of medicine materials.
