Objective: Taking Dendrobium officinale(Dendrobium officinale Kimura et Migo)as the research object,to analyze the morphological characteristics,the quantitative microscopic characteristics and index components in(baorig on)the different growing years and experimental treatment.And based on the quantitative analysis of microscopic characteristics to explore the evaluation method and technology to provide technical support and experimental basis for the identification and development of Dendrobium officinale.Methods: 1.Experiment group: The same kind of Dendrobium candidum is cultivated in 3 modes,that it is including namely control group(Masking degree 70%),physical group(Masking degree 50%)and Chemical group(Spraying plant Growth Hormone GA3).2.Morphological characteristics analysis: A direct measurement method is used to measure its morphological characteristics.3.Quantitative microscopic characteristics analysis: Using paraffin section method and NIKON Eclipse Ni microscope to measure the microscopic characteristics in transverse section of Dendrobium officinale.4.Index component analysis: Based on the China Pharmacopoeia of 2015 edition,detecting the content of polysaccharides and mannos in Dendrobium officinale.5.Data analysis: Using SPSS23.0 data analysis software to explore the relationship of Index component with microscopic characteristics and morphological characteristics.Results: (1)The results of morphological analysis show that there are differences in the height of plants,number of leaves,number of nodes,stem coarse,fresh weight,dry weight and folding rate of Dendrobium officinale in different growing years in the same treatment group(Physical group,Chemical group,Control group).Comparing the different treatment groups in the same growing year,the length of the internodes and the stem in the physical and chemical groups are higher than those in the control group,and the average stem size of the samples in the physical group is the largest in these three years.The first year is 5.56 cm,the second year is 5.13 cm,third year is 6.00 cm;the number of leaves in the physical group for three years is higher than that in the control group,and the height of plants and fresh weight of the samples in these three years are higher than those in the control group.(2)The results of quantitative analysis of microscopic characteristics show that in the same treatment group,the percentage of vascular bundle in cross section and the percentage of hard structure occupied area are as following: The second year is bigger than third year,and the first year is the smallest;in the second year samples,spraying of plant hormones can reduce the vascular bundle and the hard structure content of the stem cross-section,the intensity of the light can increase the content of the bundle and the hard structure increased;in the chemical group,the second year samples have the lowest percentage of vascular bundle area,and the average of hard structure are 1.88% and 1.67% respectively.(3)The results show that there are significant differences in mannose content of different treatment groups(p<0.05)in the same growing year.The contents of polysaccharide and mannose in the second year samples in three treatment group have reach the requirement of "Chinese Pharmacopoeia",among them,the content of mannose in the second year samples is highest,the content is 25.37%.As for the chemical group,the second year samples is highest,the content of polysaccharide reaching 38.13%.(4)The correlation analysis show that in the correlation analysis between morphological characteristics and index components,there are positively correlated in different treatment groups of polysaccharide and mannose content with the number and dry weight of Dendrobium officinale(R between 0.721-0.999);In the correlation analysis of microscopic characteristic quantification and index component,there are negatively correlated with different treatment groups polysaccharides.And the cross-sectional area percentage of the transverse section of Dendrobium officinale,the hard structure of the cross section and the percentage of cross-sectional area(R between 0.793~1.000).Conclusion: By comparing the quantitative microscopic characteristics,morphological characteristics,polysaccharide and mannose content,the quality differences of Dendrobium officinale in different growth years are obtained,which provided scientific basis for the identification and development of Dendrobium officinale.And the methods of quantitative data in microscopic characteristics could be used to determine the quality of medicinal materials,and provide new technical means for the identification and quality control of Dendrobium officinale.Innovation:1.It is the first time to make a research on the correlation between microscopic characteristics and quality of Dendrobium officinale.And the way using the experimental data provides the relationship of microscopic characteristics and the content of polysaccharides and mannose in Dendrobium officinale.2.The microscopic identification method and microscopic image processing software are used to analyze the microscopic characteristics in the transverse section,which innovating the technology and method of microscopic identification of Dendrobium officinale.3.The technology of the correlation analysis of the morphological characteristics,microscopic characteristics and index components of Dendrobium officinale is provided,and the scientific
