Objective:To explore the role of TLRs/NF-κβsignaling pathway and the effects of ecdysterone inOLP.Methods:1. HOK cells were culturede in subculture.2. We established a correlated culturedmodel of HOK cells in vitro to induce the immune response circumstance of OLP lesions byLPS,and tested the expression of NF-κβp65,TLR4and TNF-α mRNA by Real timeRT-PCR.3. We used the LPS with the concentration of10ug/ml to treat the HOK cells for6hours,and then tested the expression of NF-κβp65,TLR4and TNF-α mRNA by RT-PCRbefore and after the treatment of EDS.Results:In in-vitro-cultured OLP model, the expression of NF-κβp65、TLR4and TNF-α mRNAwas significantly upregulated after the treatment of LPS with10ug/ml final concentrationand for6hours.EDS could distinctively downregulated the expression of NF-κβp65,TLR4and TNF-α mRNA induced by LPS,and the effect of EDS was strongest with100ug/mlfinal concentration(P<0.01).
