Objective:In order to investigate the relationship of single nucleotide polymorphisms rs7903146, rs26770, rs5219 of TCF7L2ã€ARL15ã€KCNJ11 gene with T2DM and MA in Chinese-Korean in Yanbian. Methods:23 T2DM trios and 90 families of Yanbian area were collected;Using Snp-LDR method for detection of genotype;Detected ARL15 adiponectin(PA) and insulin(INS) levels by enzyme-linked immunosorbent assay (ELISA) in all samples; Detected fasting blood glucose (FPG), total cholesterol (TC), triglyceride (TG), high density lipoprotein (HDL-C) and low density lipoprotein (LDL-C) by automatic biochemical analyze;Measurement of carotid artery intima media thickness (IMT) by color Doppler;Used Faleoner regression method to calculate the nationality of T2DM heritability in Yanbian Korean and Han; Using SNP assistant version 1.0.11 (BioData, Ltd) software to calculate the transmission disequilibrium test (TDT) and allele frequency, genotype frequency of ARL15, TCF7L2, KCNJ11 gene SNPs of T2DM trios;All samples were tested for height,weight,waist circumference, hip circumference, blood pressure, calculate body mass index (BMI) and waist to hip ratio (WHR).Results:1. Heritability estimates:the genetic rate in Yanbian area T2DM families was 51.2% of which 41.6% Korean 48.8% in han, The Han nationality is slightly higher than the Korean nationality, the difference is not statistically significant (p>0.05).The genetic of core family rate of Korean 123.4%, Han was 106.7%, The Korean nationality was significantly higher than that of the Han nationality (p=0.046).the total heritability was 110.8%;2. Results of genetic model analysis:family sample observation value of male and female were 293.6,3.11,in multi gene genetic expectations range 2.71 and 2.875. The nuclear family sample observation value of male and female 4.125,4.05 respectively, between the sexes no significant difference (P> 0.05), were often autosomal dominant expectation value range 3.676 and 4.132; 3. Resuilts of allele frequency and genotype frequency distribution:frequencies of rs26770 polymorphism GG genotype in T2DM group and the Ma group were significantly higher than that in NGT group, the differences between the group and NGT with statistical significance (Pl=0.032,OR=7,95%CI:0.855-7.312,p2=0.015, OR=8.448 95%CI:1.037-8.808), are also suffering from T2DM risk increased 7 times developing Ma risk increased times 844.8. In T2DM group and MA group performance as hidden mode.The allele frequency and genotype frequency of the other SNP were not significantly different between the T2DM group and the MA group;4. The level of ARL15 and WHR With CT genotype were significantly higher than CC individuals (p=0.05),and the level of INS and was significantly lower than that of CC. There was no significant difference between the other genotypes and other clinical parameters (p>0.05);5.On 23 core pedigrees of TDT analysis. In rs7903146,rs26770 and rs5219 three allelic gene transfer distribution differences had no statistical significance (P> 0.05), passed to the affected offspring of two different alleles probability were not deviating from 50%(P > 0.05);6.The frequency of combined genotype CC-GG-CT in MA group was significantly higher than that in group NGT (p=0.022, OR=1.897), and the frequency of T2DM group was significantly higher than that of group MA (p=0.03, OR=3.319) in group CC-AG-CT.Conclusions:l.Scattered fanmilies in T2DM first-degree relatives genetic rate was 51.2%,with polygenic inheritance;but in T2DM core pedigree genetic rate is 110.8,it is very high,is autosomal dominant inheritance,is highly correlated with the onset of genetic;2.The genotype GG of rs26770 is highly correlated with T2DM and MA, it can increase the risk of T2DM prevalence of 7 times, increase the risk of disease associated with MA 8.448 times and it is performance as hidden mode;3. The allelic gene CT of Rs7903146 locus can down INS levels increase WHR by increase ARL15 levels, and promote the risk of T2DM and the combined MA;4.The TCF7L2, ARL15, KCNJ11 genes are the micro-effect gene of T2DM combined with MA in Korean and Han nationalities in Yanbian, rs7903146, rs26770, rs5219 combined with GG-CT-CC and AG-CT-CC for the Yanbian region of T2DM combined with the risk of major vascular disease combination. |
