Objective:Malignant disease has become one of the serious harm to human health. Esophageal cancer is the ninth largest malignant diseases prevalent in many parts of the world, particularly in developing countries, treatment with surgery, but a higher rate of recurrence and metastasis, chemotherapy is not satisfactory. Therefore,the study of anti-cancer drugs to gradually shift the efficient, low natural plant.Chinese medicine studies of anti-tumor have shown great potential, especially in the development of effective anti-cancer medicine plant components. Studies have shown that the extract of walnut green seedcase can inhibit the proliferation of many kind of tumor cells in vitro. This study clarified the effects of the crude extract of walnut green seedcase and other compounds, such as:gallic acid, EtoAC, taxifolin, and catechin-7-o-apiose impact on the proliferation of esophageal cancer cells, and on the expression of CyclinD1protein. Our findings maight provid theoretical and experimental basis for the further development of traditional Chinese medicine extract for the treatment of esophageal carcinoma.Methods:The esophageal cancer cell lines including EC9706and KYSE150were treated by the extracts of walnut green seedcase, gallic acid, EtoAC, taxifolin, and catechin-7-o-apiose, and the suppression ratios of proliferation of the were measured using the MTT method. Western Blot to detect the expression of cell cycle-related proteins CyclinDl in esophageal cancer cells which after different drug treatment.Result:1. The suppression effects of the compounds on the proliferation of esophageal cancer cells KYSE150and EC9706The crude extract and extracts of four kinds of walnut green seedcase gallic acid and EtoAC had the suppression effect on the proliferation of esophageal cancer cell EC9706and KYSE150, and the crude extract, gallic acid and EtoAC can inhibit the proliferation both of KYSE150and EC9706cells compared with the control group was statistically significant difference(P<0.05).2. Western Blot Assay for CyclinDlThe finding showed that the expression of CyclinDl in KYSE150and EC9706cells were significantly decreased after treatment, by the crude extracts of walnut green Seedcases, gallic acid and EtoAC.Conclusion:The crude extracts of walnut green seedcases, gallic acid and EtoAC strongly inhibited the proliferation of esophageal carcinoma cells KYSE150and EC9706, and its mechanism may be related to arrest the cell cycle.
