" Survival with tumor," improve the clinical symptoms and improve quality of life concept attracted attention at home and abroad,Chinese medicine treatment of cancer highlight its advantages,Chinese medicine treatment of tumors growing international trend to form.Anti-tumor of Chinese medicine studies.Analysis of the chemical structure and the use of traditional Chinese medicine extraction and separation of the active ingredient is the currently exploring direction.The research is to detect the effects of GA on the proliferation,migration,invasion and cell cycle and apoptosis in the human esophageal tumor cell line KYSE150,Thereby providing theoretical tumor.Methods In our experiment,KYSE150 cell in esophageal tumor cells cultured, then,The Inhibition rate of proliferation of esophageal tumor cell KYSE150 aroused by gallic acid were surveyed using the MTT method,The effects of the migration and invasion of KYSE150 aroused by gallic acid,were analysized using the cell scratch experiments and Transwell Chamber experiments respectively.The apoptosis and the cell cycle of KYSE150 treated with GA were measured by flow cytometry.and use Western Blot.Result 1.The proliferation of KYSE150 in esophageal cancer cells:When the extracts of walnut green seedcase gallic acid concentrations were 10、20、40、80、120 μg/m L. The inhibition rates were 4.15% 、19.02%、69.64%、90.51%、93.35% respectively(F=57.48,P < 0.05). 2.The esophageal cancer cells after 24 h,the migration of KYSE150 were decreased obviously, With the increase of drug concentration,the migration of KYSE150 were decreased gradually,When gallicacid concentrations were 0、20、40、80 μg/m L.The migration rates were 77%、51%、17%、7% respectively.Its ability to inhibit the migration(F=440.49,P < 0.01). 3.The effects of invasion of esophageal tumor cell KYSE150 caused by gallic acid were measured by Transwell chamber experiments: Results is determined by counting KYSE150 which Acrossing through the membrane,The esophageal cancer cells after 48 h,the invasion of KYSE150 were decreased obviously,with concentration increasing, the Transmembrane cell count decreased,When the gallic acid concentrations were 0、20、40、80 μg/m L,The Transmembrane cell count were 124.66±5.50、87.33±8.62、51.00 ± 9.53、36.00 + 9.54 respectively,Its ability to inhibit the invasion(F=74.65,P < 0.01).4.The cell cycle of esophageal tumor cell KYSE150 treated with GA were measured by flow cytometry:With the gallic acid concentration increased,the number of the cells in G0/G1 phase was increased,the cells in S phase and G2/M phase was decreased.When the gallic acid concentrations were10、20、30 μg/m L,The proportion of cells in G0/G1 phase were 33.49%、35.80%、40.48% respectively,Showed an increasing trend,The difference was significant(P < 0.05),The proportion of cells in S phase were 46.60% 、 45.60% 、 45.29% respectively,Showed a decreasing trend,The difference was significant( P < 0.05),G2/M phase also Showed a decreasing trend.5.The explore of cell apoptosis:Gallic Acid can promote cell apoptosis,With the gallic acid concentration increased, the apoptosis rate increasing,The effects were dose-dependent obviously.. 6.The expression of cell Cyclin D1 was determined by Western blotting: to extract protein from the added walnut green seedcase gallic acid in esophageal tumor cells KYSE150,Experiments with the internal control β-actin protein,Compared with the control group,The results of image processing using Quantity one software gradation analysis,The Cyclin D1 protein of esophageal cancer KYSE150 treated with gallic acid was decreased obviously.Conclusion Gallic Acid inhibited the proliferation,migration and invasion of esophageal cancer cell KYSE150 and induced apoptosis.The mechanism of the GA-inhibited the proliferation of KYSE150 cells perhaps partly through hindering the deliverance of Cyclin D1 and arresting the cells at G0/G1 phase.
