Objective:The incidence of esophageal cancer is higher in the existing treatment,surgery and radiation therapy is the treatment of first choice, but in most cases whenend-stage belongs to confirm that there is no allegation of surgery and radiationtherapy, chemotherapy so treatment plays a very important significance.Butesophageal malignant tumor cells in chemotherapy resistance, causes of affecting thecurative effect and prognosis of patient care.Glutathione transferase (GSTs) is widely exist in organism metabolismdetoxification enzyme system, can be in glutathione thiol when combined withelectrophilic compound catalytic reaction, lead to a new generation material than theoriginal polarity, making it change solubility in water, soluble in water, easy todischarge from the body.Glutathione transferase belongs to a kind of catalyst in thebody, is associated with toxic substances or carcinogenic substance metabolismdetoxification of important enzymes.So far, a lot of research is GST-Uighur andmechanism of primary or secondary resistant malignant tumor.Astragalus as legumes Mongolia astragalus membranaceus or film pod ofastragalus root, a beneficial gas-solid table and water the effect of detumescence,detoxification, raw muscle.Can be used for the treatment of cancer, in recent years,research has shown that astragalus membranaceus compound preparation for a varietyof venomous cancer in external were to restrain the action of the growth, there are avariety of mechanisms to play a role, pharmacological action and clinical experiencewas much attention.This project use Ec9706/cDDP cellular system as a researchobject, using the quantifiable indicators, to observe whether astragalus extract canreduce cisplatin resistance of esophageal malignant tumor cells the expression ofglutathione reductase, can decrease the cell resistance effect, modern scientific basisfor the therapy of traditional Chinese medicine for our country.Methods:1, in the land of0.5ug/ml cisplatin RPMI-1640medium cultureEc9706/cDDP cell;With determined by MTT method to detect the toxic effect ofastragalus extract of cells, to find the non-toxic concentration of cells of the reversal of drug resistance test;With determined by MTT method to detect Ec9706/cDDP andno added with astragalus extract is added to cultivate of cell survival after48h, dealwith two groups of data with linear regression, work out different half inhibitoryconcentration (IC50) difference, multiples, calculate the reverse=control IC50IC50/test group.2, different cell lines can be divided into experimental and control in thetwo groups, the experimental group with astragalus extract intervention, using rt-pcrmethod to detect the expression of GST-PI two groups of cells.3, using SPSS13.0statistical software processing data, in order to+S, IC50was calculated by the linearregression, the comparison between different sets of data, using t test.Results:1, astragalus extract of Ec9706/cDDP cells have poison, associatedwith the concentration of extracts from radix astragali.1,5,10ug/ml group and thecontrol group, the inhibition rate of no difference in statistics, astragalus root extractconcentration to100ug/ml,1mg/ml group inhibition rate between differentrespectively was18.4%,29.8%compared with the control group with statisticalsignificance.So choosing the non-toxic astragalus extract concentration for10ug/mlfor reverse the concentration used.2, separate effect of cisplatin, cisplatin IC50ofEc9706/cDDP concentration is43.59ug/ml, cisplatin, and no cytotoxicity dose of10ug/ml combination of astragalus root extract, DDP IC50of Ec9706/cDDPconcentration drops to26.27ug/ml, reverse ratio of1.66.3, in10samples of group A,eight GST gene-PI has high expression level, two for the low expression level;GroupB also have10different samples, the GST gene-PI has high expression level,9forthe low expression level.By the statistical analysis shows that GST-PI geneexpression in group A and B the extent, the differences were statistically significant (P<0.05).Conclusion:astragalus extract can reverse Ec9706in experimentalconditions/cDDP cell lines, the performance of cisplatin resistance reversal effect anddose show correlation, the mechanism may have correlation with GST-lower geneexpression of PI.
