Objectives To evaluate the association between angiotensinconverting enzyme (ACE) I/D gene polymorphism,methylenetetrahydrofolate reductase (MTHFR) C-677T gene polymorphism and the ischemic stroke in the Asian populations.Methods Case-control studies published were collected from databases of Pub-Med,Embase and CNKI based on theselection criterion. Statistical analysis was p-erformed with STATA software version11.0.We calculated the pooled odds rati-o (ORs) with95%confidence interval (CI) using those extracted data.Results Thirty-nine independent publicationswith7020ischemic stroke cases a-nd9006controls were included. Of the total,21studies evaluated the associate-on. between ACE I/D polymorphism and ischemic stroke,18studies evaluated the association between MTHFR C677T polymorphism and ischemic stroke. F-or the ACE I/D polymorphism, the carriers of DD genotype was1.60times higher risk with ischemic stroke than the other variant genotypes (OR=1.60,95%CI:1.24-2.06,P<0.01); the D allele suggested asignificant association with is-chemic stroke (OR=2.21,95%CI=2.06-2.38). Meta-regression indicated that the gender, geographic region (China Japan Korea India Malaysia)had no associatio n with ischemic stroke both for the MTHFR C677T polymorphism and the AC-E I/D polymorphism. Potentialpublication bias might exist in the studies of M-THFR C677T polymorphism, but not for the studies of ACE I/D polymorphis-m (P>0.05). Correction for this bias using a formal statistical method did not materially alter thecombined risk estimate. The resultof sensitivity analyses indi- eating that the pooled estimate was robust and notinfluenced by a single study.Conclusion The results of this meta-analysis strongly suggest that theACE I/D polymorphisms significantlycontribute to the risk of ischemic stroke and the Tal-lele of MTHFR C677T was significantly correlated to ischemic stroke suscepti-bility in the Asian populations.
