Object: To study whether the single nucletide polymorphisms(SNPs) of ATP-binding cassette transporter 1(ABCA1) gene and 3-hydroxy-3-methylglutaryl coenzyme A (HMG-COA) reductase gene are associated with cerebral infarction(CI) in Chinese,and furtherly discuss the pathogenesis of cerebral infarction.Method: Using polymerase chain reaction technology and restriction fragment length polymorphism technique to analysis the target fragments of polymorphic loci of ABCA1 gene and HMG-CO A reductase gene.By contrust in 90 patients and 100 controls,we detected the SNPs of ABCA1 gene and HMG-CO A reductase gene. The frequencies of allele and genotype of SNPs of ABCA1 gene and HMG-CO A reductase gene were Statistically analyzed by statistical software package SPSS13.0Result:1 The gene distribution of Hardy-Weinber genetic equilibrium test (P> 0.05) consistent with the genetic balance.2 The polymorphism genotype (P = 0.047) and allele frequency (P = 0.016) of the SNP of I883M in ABCA1 gene has a significant difference between 90 patients and 100 controls. I allele frequency of cerebral infarction was significantly higher than that of healthy controls (74%, 61.8%), may be related to cerebral infarction.3 The II genotype of I883M in ABCA1 gene was significantly higher than that of healthy controls (8.9%,4%), may be related to cerebral infarction.There were no statistical significantly different between the two groups(p=0.234).4 There hasn`t significant difference between 90 patients and 100 controls in the allele frequency (P = 0.016) of the SNP of HMG-COA reductase gene. A allele frequency of cerebral infarction has no significantly higher than that of healthy controls (51.7%, 49.5%), may be not related to cerebral infarction.5 There were significantly difference of the age(p=0.027), plasma total cholesterol (TC), triglyceride (TG) (P<0.001), high-density lipoprotein (HDL) (P < 0.001),GLU(p=0.003), and drinking history HDL(P=0.0482)and hypertension (P<0.001) between cerebral infraction and healthy controls.But there was no significantly difference of the gender (p=0.027), plasma total cholesterol (TC) (p=0.208) between cerebral infraction and healthy controls6 When taking ABCA1 and the HMG-CO A reductase gene polymorphism locus genotype, age, gender triglyceride (TG), plasma total cholesterol (TC), high-density lipoprotein (HDL), and drinking history and other potential risk factors into consideration, the Logistic regression analysis didn`t show they may be risk factors for cerebral infarctionConclusion:These suggested that ABCA1 I883M may be related to cerebral infarction in HeBei province Han people, and HMG-CO A reductase gene was not the risk factors for cerebral infarction.
