Objective: Though analyze the construction of sputum bacterial flora whichcomes from patients with COPD, to study the relationship between the changes ofsputum bacterial flora and the progression of COPD. Respiratory tract flora will benew target for clinical prevention, diagnose and treatment of COPD, this will expandway of thinking of personal treatment of disease of respiratory system.Methods: Collect sputum sample of clinical patients with COPD according to thesputum bacteriology inspection standard operating procedures, extract total bacterialDNA from sample, PCR technology and primer was used to expand16S rRNA ofsample. DGGE was used to separate expanding production the Map of sputumbacterial species structure was obtained, meaningful DNA banding was cut and cloningsequencing. Real-time PCR was used to analyze the changes of klebsiella pneumoniae,pseudomonas aeruginosa and streptococcus pneumoniae in sputum of patients withCOPD.Results: There is significant difference between COPD group and health group ofDGGE map of sputum bacterial (P <0.05).Contrast with normal group, the number ofklebsiella pneumoniae, pseudomonas aeruginosa and streptococcus pneumoniae inCOPD group were significant increased.Conclusion: Combined DGGE and Real-time PCR will helpful to evaluate thechanges of sputum bacterial flora. The construction of sputum bacterial flora andnumber of particular sputum bacterial were significantly changed. There certaincorrelation between the changes of sputum bacterial flora and the progression.
