Objective: To explore the effect and mechanism of artesunate (ART) on anti-humancervical cancer cells.Methods:1. Clinical research: The population of CD4+CD25+FoxP3regulatory T cells (Treg)in peripheral blood from60cervical cancer patients and30healthy volunteers weredetected by flow cytometry.2. Cell experiments: FoxP3mRNA and protein expression in Jurkat T cells stimulatedby the supernatants of CaSki and Hela cells treated with ART was determined usingquantitative polymerase chain reaction (qPCR) and western blot analysis.3. Animal experiments: In vivo antitumor activity was investigated in mice withcervical cancer by the subcutaneous injection of various concentrations of ART. Thepopulation of CD4+CD25+FoxP3regulatory T cells (Treg) in peripheral blood from micewas detected by flow cytometry,and the change of tumor volume was observed.Results:1. The population of CD4+CD25+FoxP3Treg cells in the peripheral blood wassignificantly higher in cervical cancer patients than healthy volunteers (P<0.05).2. The supernatants of CaSki and Hela cells treated with ART lowered the expressionof FoxP3in Jurkat T cells.3. ART was capable of inhibiting orthotopic tumor growth, which correlated with adecrease in the percentage of Treg cells in mice with cervical cancer.Conclusion:1. The increase of CD4+CD25+FoxP3Treg cells population in the peripheral bloodfrom cervical cancer patients indicates that CD4+CD25+FoxP3Treg cells population isassociated with cervical cancer development. 2. ART was capable of inhibiting orthotopic tumor growth, which correlated with adecrease in the percentage of Treg cells in mice with cervical cancer.3. ART effectively inhibited FoxP3expression in T cells through cervical cancer cellsand led to the inhibition of tumor growth.
