Objective To investigate the relationship between the expressions of Smad2,Smad4andSmad7and renal Chronic allograft nephropathy(CAN). We expected detection of expression ofSmad proteins in urine is a method of noninvasive diagnosis of CAN.Methods Divide the60Kidney transplant patients into2groups, the CAN group is chronictransplant kidney nephrosis patient(30cases); Control group is non-chronic transplant kidneynephrosis patient(30cases), We take two group of morning center-section urine for study。Examines the CAN group and control group patient expression of Urinary Smad2, Smad4, theSmad7protein with enzyme immunity technology (Elisa),Examines the CAN group and controlgroup patient mRNA expression of Urinary Smad2,Smad4, the Smad7protein with Real-Time PCR.Results With Elisa examination, CAN group expression of Urinary Smad2, Smad4, theSmad7protein levels are differences with the control group (P<0.05). RT-PCR showed that CANgroup of urine samples and transplantation in patients with Renal Biopsy in the mRNAexpression of Smad2and Smad4is significantly higher in patients with control group, rather thancontrol group urine samples and transplantation in Renal Biopsy tissues mRNA expression ofSmad7significantly higher than CAN group.(P<0.05).Conclusions Through our research, CAN group expression of Urinary Smad2and Smad4Proteomic were significantly higher in patients with the control group, while expression of Smad7is significantly lower than the control group, therefore we believe that urine Smad2, Smad4andSmad7protein combined diagnosis CAN have a higher degree of sensitivity and specificity. Webelieve that it is a more convenient method of non-invasive diagnosis of chronic allograftnephropathy. It has provided very valuable reference in clinical diagnosis for the clinician and areliable biological indicators for follow-up kidney transplants the patient.
