OBJECTIVE: To evaluate the expression and significance of Bmi-1, Mel-18and Notch1protein in invasive ductal carcinoma.METHODS: Expressions of Bmi-1, Mel-18and Notch1protein in68cases ofbreast IDC,32cases of breast fibroadenoma and30cases of normal breast tissuewere detected by immunohistochemistry(IHC), and then their relationship withrelated clinical information in breast IDC were analyzed, We also evaluated thecorrelations of their expressions. All data were analyzed using the SPSS16.0softwarepackage.RESULTS: The positive expression rate of Bmi-1protein in breast IDC, breastfibroadenoma and normal breast was75.0%,34.38%and16.7%, There weresignificant differences among three groups comparison(P<0.05); The positiveexpression rate of Mel-18in breast IDC, breast fibroadenoma and normal breast was26.5%,59.4%and73.3%, The positive expression rate of Notch1in breast IDC,breast fibroadenoma and normal breast was80.9%,53.1%and33.4%, they bothsignificant differences among three groups (P<0.05). The protein expression level ofBmi-1, Mel-18and Notch1was correlated with lymph node metastasis, TNM andhistological grade(P<0.05), it was not correlated with the estrogen acceptor(ER),progestogen acceptor(PR) and the age does not have the relevance(P>0.05). Theexpression of Bmi-1was positive related to that of Notch1in breast IDC, they bothhigh expression in human breast cancer. The expression of Mel-18was lowexpression in breast IDC, it was negatively related to that of Mel-18..CONCLUSION:1. Bmi-1and Notch1protein were highly expressed in breast IDC, and were closely related with lymph node metastasis, the degree ofdifferentiation, which suggested that Bmi-1and Notch1gene play important roles inbreast IDC occurrence, development and progression.2. Bmi-1protein was highly expressed in breast IDC, but Mel-18protein waslowly expressed in breast IDC, which suggested that Mel-18might affect theprogression of breast IDC via down-regulating Bmi-1protein expression.
