Objective: To explore the significance of Notch1 and Snail in the development of breast carcinoma, the mRNA expressions of Notch1, N1 IC and Snail were analyzed in adjacent normal breast tissue(ANBT), atypical ductal hyperplasia(ADH), ductal carcinoma in situ(DCIS) and infiltrating ductal carcinoma(IDC). Methods: The mRNA expressions of Notch1, N1 IC and Snail were measured by qRT-PCR from 80 IDC, 20 DCIS, 15 ADH and 25 ANBT tissues. The correlations between three gene mRNA expression were further analyzed. The correlation between the three genes mRNA expression with clinicopathological features in IDC were analyzed, and relativity between the three genes expression combined with the previous findings of protein expression. Results:(1)Notch1 m RNA expression in ANBT(0.55±0.26), ADH(0.81±0.34), DCIS(1.79±0.62) and IDC(2.42±0.99) were gradually increased, the significantly statistical differences(P=0.000, P=0.000) while IDC and DCIS groups compared with ANBT group respectively, The positive correlation between Notch1 mRNA expression and histological grade(P=0.000), lymph node metastasis(P=0.000) and TNM stage(P=0.02), Notch1 mRNA and protein expression(previous results) showed a significant positive correlation(r=0.404, P=0.000);(2) The increasing tendency of N1 IC mRNA expression in four groups is consistent with Notch1 expression(ANBT:0.70±0.32,ADH:1.05±0.44, DCIS: 2.09±0.75, IDC: 2.19±0.97), there were significantly statistical differences(P=0.000, P=0.000) while IDC and DCIS compared with ANBT, N1 IC mRNA expression positive correlated with histological grade(P=0.000), lymph node metastasis(P=0.000) and TNM stage(P=0.03), N1 IC mRNA and protein expression showed a significant positive correlation(r=0.332, P=0.003);(3)Snail mRNA expression were increasd in breast tissues(ANBT:0.55±0.36,ADH:1.06±0.43,DCIS:2.18±0.77,IDC:2.51±0.98), there were significantly statistical differences(P=0.000, P=0.000, P=0.001) while IDC, DCIS and ADH compared with ANBT, furthermore, The positive correlation between Snail mRNA expression with lymph node metastasis(P=0.001), TNM stage(P=0.000), The positive correlation of Snail mRNA and protein expression(r=0.217, P=0.054) was no statistical significance.(4) Three genes mRNA expression in four breast tissues(IDC: r=0.235,P=0.036; DCIS: r=0.554,P=0.032; ANBT: r=0.522,P=0.018) showed a significant positive correlation. Conclusions:(1) The mRNA expressions of Notch1, N1 IC and Snail were gradually increased during breast canceration, suggesting a potential role in the initiation of breast cancer.(2) expressions of three genes positively associated with lymph node and TNM stage, Notch1 and N1 IC expression positively associated with histological grade, its may be promote the progression of breast cancer patients.(3)Notch1-N1IC-Snail expression significantly positively related in four breast tissues may play a synergy role in the metastasis of breast cancer.
