[Background]:PHD2(prolyl hydroxylase domain2, proline hydroxylase2) is alsocalled EGLN1(egg laying about nine1), as a family of proline hydroxylase (PHDs) isthe most important oxygen sensors, oxygen dependence catalyzed by hypoxiainducing factor (HIF) proline residues of specific hydroxyl reaction occurs, thusmediated degradation oxygen induced factor, have an effect on hypoxia inducingfactor transcription activity,may improve gastric carcinoma-related desmoplasticresponse.[Purposes]:1、To investigate the correlations of expression of PHD2protein between gastriccarcinoma-related desmoplastic response and clinicopathologic feature;2、 To explore the correlations of nucleotide mutation of PHD2in gastriccarcinoma-related desmoplastic response.[Methods]:1、Screening of690cases of gastric cancer in2007-2012. Divided into four groupsaccording to the extent of carcinoma-related desmoplastic response under themicroscope. In turns, scirrhous gastric cancer that promoting fiber reactionsignificantly (156cases), the intermediate like scirrhous gastric cancer (182patients), intermediate with medullary growth patterns of gastric cancer (188cases), the myeloid growth of gastric cancer with rare profibrogenic (164cases).2、Detected the level of PHD2protein in the four groups samples byimmunohistochemical and statistical analysis the expression of PHD2in gastriccancer, and clinical pathological features, infering relationships of PHD2and ggastric carcinoma-related desmoplastic response. 3、Detection gastric cancer two known mutations of PHD2gene by LDR-PCR in the690patients. Selecting randomly50cases of adjacent relativelynormal gastric mucosa as normal controls in four groups of gastric cancer.[Results]:1、The expression of PHD2in scirrhous gastric cancer that promoting fiber reactionsignificantly is lower than any other three groups;2、Vessel tumor thrombus and cancer cells in lymph nodes metastasis have positiveexpression of PHD2;3、The worse histological, the higher TNM staging, with lymph node metastasis owelower positive rate of PHD2;4、PHD2gene SNPs, c.471G>C(p.Q157H)、c.740C>G(p.S247W)allele andgenotype frequencies no difference between the four groups of different fiber-reactivegastric tumor and paraneoplastic relatively normal gastric mucosa, suggesting nosignificant correlation between the base changes and occurrence of tumor; four groupsof different fiber-reactive gastric tumor tissue PHD2gene SNPs,c.471G>C(p.Q157H)、c.740C>G(p.S247W)allele and genotype frequencies no difference,suggesting two pointsbase changes and cancerous profibrogenic reaction was nosignificant correlation.[Conclusion]1、 PHD2associated with the biological behavior in gastric cancer, the strongestcarcinoma-related desmoplastic response of scirrhous gastric cancer wassignificantly lower than any other three groups about PHD2positive rate.2、 The expression of PHD2involving in the evolution of gastric cancer biologicalbehavior.3、 There’s no PHD2gene point c.471G>C(p.Q157H)、c.740C>G(p.S247W)mutation detected in gastic cancer.
