Objective: To investigative the relationship between single nucleotidepolymorphism (SNP) of codon72of the P53gene (Arg72Pro) and occurrences ofgastric cancer in QingHai, China.Method:Polymorphisms of the genotype of P53codon72were identified byPolymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP). The test group is consisted of genomic DNA samples from123patients whilethe control group is consisted of genomic DNA samples from84healthy individuals.Representative samples in the SNP loci diversity were tested by Hardy-Weinbergequilibrium test. SPSS15.0software package was used to perform statisticalanalysis including χ2test and Fisher’s exact probability analysis in SNP loci genefrequency and genotype frequency distribution in control group as well as the testgroup.Results:In QingHai province, the frequencies of Arg/Arg, Pro/Arg and Pro/Progenotypes of the P53codon72polymorphism were21.4%,50%and28.6%in thecontrol group (healthy individuals), while the respective frequencies were36.5%,47.2%and16.3%in the test group (gastric cancer patients). There is a statisticaldifference (p<0.05) in the P53gene Arg/Arg genotype, which was also associatedwith pathologic grade at the risk of gastric cancer and lymph node metastasis. Thepolymorphism, however, was not correlated to age, gender, ethnicity, tumor location,depth of invasion and Borrmann type.Conclusions:There is a significant relationship between the frequencies ofArg72Pro polymorphism of the P53gene and the risk of gastric cancer in the QingHaiprovince of China, indicating that the P53gene Arg72Pro polymorphism plays an important role in the incidence of gastric cancer. Therefore, the P53gene Arg72Propolymorphism would be promising as an indicator of the gastric cancer as well as thetherapeutic target if its functions and mechanisms could be further investigated.
