Banxia-Houpu Decoction from the Han Dynasty Zhang’s "Golden Chamber”, theprescription by the Pinellia, Magnolia, Poria, Ginger and Perilla leaves composition,there is qi stasis, phlegm Jiang-ni efficacy, clinical application can be found sodepressed mood depression into comfortable pleasant, it is now used in the treatmentof depression. Depression, also known as major depressive disorder, with significantand persistent low mood as the main clinical features are the main types of mooddisorders, depression is a medical category. The etiology and pathogenesis ofdepression is very complex, may cause long-term depression in patients with a varietyof biochemical abnormalities. By studying the changes in the experimental rat urinemetabolites and serum explore metabolomics difference among the model group,treatment group and the control group of rats. Study is divided into the followingsections:(1)Established depression model of rats. The model group conducted the modelprogram of depression, for a period of24days. The result show that the weight of themodel group growth is slow, the immobility time increased and sucrose preferencerate decreased, when compared with the control group. It suggests that the model iseffective. Serum and urine metabonomics study found, the implementation modelafter the procedure, small molecule metabolites relate to amino acids and energymetabolism appeared abnormal, infer its possible mechanism, found very similar andpathological mechanism of depression in patients with depression, showed that themodel was successful.(2) Study on the resistance effect of Banxia Houpu Decoction on depression in rats.This experiment has administered group, model group and control group. Exceptingthe control group, the remaining two the remaining two groups were made toimplement the model program of depression for24days, the next day to giveBanxia-Houpu Decoction of water extract. The results show that the treatment groupweight growth rate higher than that of the model group, immobility time value andsucrose preference rate mediated the control group and the model group. The urine and serum samples metabolic studies found that after treatment, the abnormal changesof depressant rats, such as succinic acid, citric acid,2-oxoglutarate, tryptophan andphenylalanine are effectively reversed, the analysis of these metabolites in vivomechanism of formation and elimination pathways and found that they produceabnormal changes and depression there is a certain correlation, indicatingBanxia-Houpu Decoction has an antidepressant effect.3) Study on the antidepressant effect dose-effect relationship of the Banxia-HoupuDecoction water extraction. This experiment has control group, model group andthree different doses of the treatment group. Excepting the control group, theremaining four groups were made to implement the model program of depression for24days. The results showed that the value of the immobility time of the high-dosegroup is less than the middle-dose and the low-dose group, the value of weight andsucrose preference is greater than the middle-dose and the low-dose group, initiallyconfirmed with the dose increased, the antidepressant effect of strengthening.Metabonomics research on urine and serum found that the high-dose groupsignificantly due to depression caused by the reversal of energy and amino acidsabnormalities, the low-dose group almost irreversible metabolic abnormalities suchdepressive state. Indicating that the antidepressant effect of Banxia-Houpu Decoctionhas a certain dose-effect relationship.Antidepressant-related biomarkers of Banxia-Houpu Decoction in this experimentwere assigned on the antidepressant effect relationship were discussed. The studyshowed that the use of metabolomics technology Banxia-Houpu Decoctionantidepressant effects research has practical, reliable advantages.
