Purpose:To study the effect of Supplemented Banxia Houpu Decoction on behavior, thymus structure and phagocytic function in despair and depression mice, to discuss the decoction's antidepressant effect and its mechanism.Material and method:①Forty female Kunming mice(18-22g) in clean grade were selected, which were divided into 4 groups randomly:control group, model group, Traditional Chinese Medicine group and foreign medicine group,10 mice in each group.②Except the control group, stressed the mice by forced swimming test(FST).③1 hour before the FST, gave Supplemented Banxia Houpu Decoction to the traditional Chinese Medicine group; gave fluoxetine hydrochloride to the foreign medicine group; gave normal saline to model group.④Open-field test were taken to observe the behavior changes in mice; the thymus index were calculated by the ratio of thymus weight and body weight; thymus specimens were fixed for observing microstructure and ultrastructure changes by light microscope and electron microscope; thymic lymphocytes were cultured in vitro, using MTT assay to test the changes in proliferation of mice thymic lymphocytes; carbon clearance test were taken to calculate the clearance index (K) and phagocytic index (a).⑤The data was expressed by x, statistical analyzing with SPSS11.0.Results:1. Behavioral comparison:The staying time in center grid of model group was significantly longer than control group, and the times of modified was significantly reduced(P<0.05), Compared the control group and foreign medicine group, there was no significant difference with Chinese medicine group.2. Thymus index changes:Compared with control group, the thymus index of model group decreased obviously (P<0.05), Compared the control group and foreign medicine group, there was no significant difference with Chinese medicine group.3. Thymus structure changes:The thymus of model group mice were small, not gloss and hard to peel. Microscopic structure showed that thymocytes arrangement relative sparse, gaps grew. Ultrastructure showed that the nucleus had irregular shapes, get small, arranged scattered, the number of them were significantly reduced, some cells showed buckling of the membrane shape, boundaries disapp-eared, mitochondria significantly increased, the cristae were disorganized, with the majority of swelling, vacuolization. Mitochondria of Chinese medicine group are a slight swelling, vacuolization, membrane and nucleus did not change significantly, it had clearly improved compared with model group.4. Thymocyte proliferation:The model group significantly reduced the rate of thymocyte proliferation(P<0.05), Compared the control group and foreign medicine group, there was no significant difference with Chinese medicine group.5. Phagocytic function:Compared with control group, the clearance index (K) and phagocytic index (a) of model group decreased obviously (P<0.05), Compared the control group and foreign medicine group, there was no significant difference with Chinese medicine group.Conclusion:1. Supplemented Banxia Houpu Decoction can construct the cognitive ability of behavioral despair and depression mice.2. Supplemented Banxia Houpu Decoction can protect the thymus structure of behavioral despair and depression mice.3. Supplemented Banxia Houpu Decoction can improve thymus index and prolifera-tive activity of thymocytes of behavioral despair and depression mice.4. Supplemented Banxia Houpu Decoction can improve the phagocytic function of behavioral despair and depression mice.
