Expression Of MicroRNA-203 In Plasma And Its Significance In Diagnosis Of Gastric Cancers

Objective:Traditional tumour marker was detected in the diagnosis of gastric cancer has a wide application, however, has not yet to find a more specitific and more sensitive marker in diagnosis of cancers. As for the tumor's occur and vest have been studied deeply,the abnormal expression of the microRNA in tumor tissues bring the hope for early diagnosis of gastric cancer, at the same time, the latest research shows microRNA-203 and gastric cancer of the occurrence, development has a definite correlation. However, there are more restrictions in the source of pathological specimens,the detection of the microRNA-203 in blood become the new direction of gastric cancer's diagnosis. This issue through detecting the experssion of microRNA-203 in gastric cancer patients'blood, to investigating microRNA-203 in the diagnostic value of gastric cancer.Methods:In this study,30 plasma samples collected from the 30 patients of gastric cancer,and 10 plasma samples collected from adult normal human, which obtained from Shanxi tumor hospital between June 2008 to June 2009.All patients diagnosed by two seasoned pathological doctors,according to the classification standard for diagnostic or classification in the world health organization of gastric cancer. The expression of mircoRNA-203 in 30 cases of gastric cancer and 10 cases of control were detected by Real-time PCR.The relationships with the expression of mircoRNA-203 and clinical pathology parameters also analyzed. Receiver operation characteristic (ROC) curve analysis was performed to determine the specificity and sensitivity of mircoRNA-203 as a diagnostic biomarkers in Gastric Cancers.Results:The experssion of mircoRNA-203 in blood has remarkable differences in gastric cancer group and control group. MircoRNA-203 has significantly low expression in the blood of gastric cancer group. Low expression of miR-203 was correlated with tumor size parameters of the invasion and lymphatic,remarkable depth, transfer lines, pTNM stage; Invasion into vessels, lymphatic metastasis, Histologic grade.The area under curve(AUC) degree was 0.97 have a high accuracy.Conclusions:Low expression of the microRNA-2003 in blood of gastric cancer shows that it may act as a tumor suppressors in the pathgenesis of gastric cancers. At the same time miR-203 palys a regulate role in the progression of gastric cancers The area under curve(AUC) degree was 0.97,it shows a higher accuracy in diagnosis of gastric cancers by detecting the expression of microRNA-203 in blood.
