Objective: 1.Selecting the intraoperative tumor tissue,adjacent tissue of patients and mammary tissue of healthy women.2.Detecting the expression of miR-155 in serum between breast cancer patients and healthy controls by real-time quantitative polymerase chain reaction(real-time q PCR)analysis.3.Measuring the methylation level of intraoperative tumor tissue,adjacent tissue of patients and mammary tissue of healthy women by MSPCR.4.Combing clinical data to anylysis he relevance between miR-155 and clinical pathology data.Results:1.There is significantly difference in miR-155 between breast cancer patients and healthy controls(p<O.05),and miR-1 55 is up-regulated in serum of breast cancer.2.The expression level of miR-155 in patients with positive lymph nodes was up-regulated compared with that in patients with negative lymph nodes(P<0.05).The expression level of miR-155 in stage Ⅲ breast cancer was up-regulated compared with that in stage I&II breast cancer(P<0.05).The expression level of miR-155 in patients with positive ER and PR was down-regulated compared with that in patients with negative ER and PR breast cancer.3.Methylation rates of miRNA155 gene in cancer group and control group were 30.95 and 37.5%(f=10.614,P<0.05).Conciusion:1.MiR-155 is up-regulated in breast cancer in serum of breast cancer patients compared with healthy controls.MiR-155 may become a novel class of biomarkers for early diagnosis of breast cancer.2.MiR-55 in serum of breast cancer is relevant with clinical stage molecular subtypes.The relative expression of late clinical stage,negative hormone receptor is higher;otherwise lower.3.Lower methylation rate of miRNA155 gene occur in breast cancer.Abnormal methylation of miRNA155 gene may promote breast carcinogenesis.
