| Objective:1. To experimental study the expression of Vimã€CK20ã€CD44v6in the tissue ofnormal cervical glandular epithelium,cervical adenocarcinoma in situ,cervicaladenocarcinoma/adenosquamous carcinoma, to discuss the relationshipbetween these three protein and clinical pathological parameters of cervicaladenocarcinoma/adenosquamous carcinoma.2. To retrospective analysis HR-HPV prevalence in cervical adenocarcinoma/adenosquamous carcinoma,and discuss the relationship between HR-HPVinfection and clinical pathological parameters of cervical adenocarcinoma/adenosquamous carcinoma.3. To statistical analysis the relevance among the expression of Vim,CK20,CD44v6in cervical adenocarcinoma/adenosquamous carcinoma,whetherHR-HPV infection is associated with the expression of these three protein inthe tissue of adenocarcinoma/adenosquamous carcinoma.Method:1. Utilize immunohistochemical technique to detective the expression ofVim,CK20,CD44v6in paraffin-embedded tissue of normal cervical,retrospective analysis HR-HPV prevalence in cervical adenocarcinoma/adenosquamous carcinoma.2. Collect the clinicopathologic data of clinical-experimental group in the researchand follow–up all of these patients.3. Analysis the relevance among the expression of Vim,CK20,CD44v6and thecondition of HR-HPV infection.Result:1. There no expression of Vim,CK20were observed in tissue of normal andadenocarcinoma insitu,but26%,21.8%in adenocarcinoma/adenosquamouscarcinoma. The differences between adenocarcinoma/adenosquamous carcinoma and normal, adenocarcinoma in situ of the study were striking(P<0.05),and nodifferences between normal and adenocarcinoma insitu (P>0.05).Expression ofCD44v6was ratchet up from nomal to adenocarcinoma in situ and adenocarcinoma/adenosquamous carcinoma, the positive rates was20.00%,35.00%and82.80%respectively. There are significant differences between the two groups in terms of the statisticalresult of Adenocarcinoma/adenosquamous carcinoma and normal, adenocarcinoma insitu, statistical result was χ2=31.13,P=0.00<0.05,χ2=19.20,P=0.00<0.05respectively,and no significant differences between normal and adenocarcinoma in situ,statistical result was χ2=1.13,P=0.29>0.05.2. The expression of Vim in adenocarcinoma/adenosquamous carcinoma isclosely related with clinical pathological parameters of lymph node metastasis andvascular invasion(P=0.00), prognosis(P=0.01).CK20is closely related withpathological grading (P=0.00), lymph node metastasis and vascular invasion(P=0.00), CD44v6is closely related with tumour size(P=0.03), depth of invasion(P=0.00), pathological grading.3. Empirical evidence has shown positive correlation between CK20andCD44v6(R=0.24,P=0.02<0.05),but there are no correlation between Vim andCK20(R=0.21ã€P=0.12>0.05),and the same to Vim and CD44v6(R=0.125ã€P=0.09>0.05).HR-HPV also has no correlation with the expression ofVim,CK20,CD44v6,there statistical result was R=0.13,P=0.09>0.05, R=0.01,P=0.11>0.05, R=-0.02,P=0.10>0.05respectively.4. The positive rate of HR-HPV infection in the patients of experimental groupwas78%, adenocarcinoma and adenosquamous carcinoma positive rate were73.81%and85.37%respectively.All the clinical pathological parameters has norelated with HR-HPV.Conclusion:1. Following with carcinogenesis of cervical normal glandular epithelium,expressionofVim,CK20,CD44v6inthetissueof cervix arechanging.2. The expression of Vim in adenocarcinoma/adenosquamous carcinoma is closely related with micrometastasis,and would be expected to be associated withpoor outcome.CK20is closely related with invasive and micrometastasis.Highexpression of CD44v6might increase the level of invasive and malignancy.HR-HPV infection has no association with the expression of these three protein in thetissue of adenocarcinoma/adenosquamous carcinoma.3. The positive rate of HR-HPV infection in the patients of adenocarcinoma/adenosquamous carcinoma is high,and compared to adenocarcinoma patients,infection rate in adenosquamous ones is much higher. |
