Smilacina japonica A.Gray is a kind of edible perennial herb with effects of tonifying,immune regulation and mammary gland inflammation resistance etc. The current researches onSmilacina japonica at home and abroad mainly focus on the identification and analysis of itschemical composition and extraction process of amino acid from Smilacina japonica has notbeen reported in literature so far. For the sake of better development and utilization ofSmilacina japonica, extraction, decolorization and purification of free amino acids fromSmilacina japonica were studied systematically in this paper. Finally, the application of aminoacid in cosmetics was preliminarily discussed.Free amino acids from Smilacina japonica were extracted by ultrasonic assisted extractionmethod. The optimal extraction condition through single factor and orthogonal experiment is:the ratio of raw material to extraction solvent1:30, extraction time30min, extractiontemperature40℃and ultrasonic power80W. Free amino acids from Smilacina japonica ofdifferent harvest period were extracted under the condition above and the results show that thefree amino acids decreased with the length of harvest time.By analyzing impacts of decolorization way, dosage of powdered activated carbon,adsorption time, adsorption temperature and rotation speed on decolorization rate and loss rateof amino acid, the optimum decoloring parameters were determined as follows:0.5g/50mL inactive carbon quantity,50min in time,80℃in temperature and120r/min in rotation speed.Under this condition, the decolorization rate can reach95.98%and the amino acid loss wasonly7.34%.The amino acid extraction liquid was purified by732cation exchange resin. The suitableexchange conditions were found through the study of the static and dynamic exchange asfollows: the sample concentration was5-6mg/mL, pH2.0with a flow rate of2.0BV/h, and thenthe column was washed by3%ammonia after5h. Collect the eluent, remove the ammonia byrotary evaporation and amino elution yield can reach87.34%. Free amino acids from Smilacinajaponica were detected and analyzed by automatic amino acid analyzer, which shows that freeamino acids from Smilacina japonica consist of16different kinds of amino acids, among which7kinds of essential amino acid are included and account for47.95%of the total amino acidcontent.The extracted and purified amino acid was added to cosmetics. As the reference, the basic Moisturizer and cream added with HA were selected. The moisturizing efficiency of the abovecream was evaluated by weight measuring. As a result, the moisture retention rate of amino acidcream was higher than basic cream, but a little lower than HA cream. The result indicated thatthe moisturizing efficiency of amino acid cream was excellent. Amino acid moisturizing creamis fine smooth with bright white glossy coating, easy to absorb, and stability and safety of it isalso in line with the corresponding national standards, indicating the cream has the possibilityof market development.
