Objective:To analyse the correlation between the deviation of tissue free amino acids of breast cancer and unaffected surrounding mammary gland and histological grading,pathologic typing,axillary lymph nodes metastasis,tumor volume and body weight,and to explore the relationship between clinical pathology and amino acids metabolic changes of cancerous tissue in patients with breast cancer.Methods:Fresh tissue samples of breast cancer and unaffected surrounding mammary gland from 20 patients with breast cancer were col lected. Seventeen kinds of free amino acids content in breast cancer and unaffected surrounding mammary gland tissue were detected by amino acids autoanalyser L-8800.Clinical data of 20 patients with breast cancer were collected.Data was expressed as mean±SD.Statistical analyses was performed using the paired t-test,correlation and multiple stepwise regression analysis with SPSS13.0,P value < 0.05 was considered statistically significant,P value < 0.01 was considered obviously statistically significant.Results:It was remarkably positive correlation between the total content deviation of seventeen kinds of free amino acids of cancerous and unaffected tissue and histological grading(P=0.001). It were remarkably positive correlation of glycine,phenylalanine,serine,aspartic acid, glutimic acid,methionine,threonine and tyrosine content deviation and histological grading(P<0.01),it were positive correlation of isoleucine, proline, arginine, histidine and lysine content deviation and histological grading(P < 0.05). There was remarkably correlation of proline content deviation and pathologic typing(P = 0.013),it was remarkably correlation of glycine content deviation and pathologic typing (P=0.002). It was positive correlation of alanine content deviation and axillary lymph nodes metastasis(P=0.021).It were remarkably positive correlation that serine, threonine and aspartic acid content deviation and tumor volume(P<0.01), it was positive correlation of glycine content deviation and tumor volume(P < 0.05).There was remarkably positive correlation of leucine content deviation and body weight(P<0.01),it were positive correlation of valine, isoleucine, histidine, tyrosine content deviation and body weight (P<0.05).Conclusions:It presented difference of tissue amino acids metabolism between breast cancer and unaffected surrounding mammary gland.There was a relationation between the majority of cancerous tissue free amino acids and histological grading,such as glycine,phenylalanine,serine,aspartic acid,glutimic acid,methionine,threonine ,tyrosine isoleucine, proline, arginine, histidine and lysine, especially glycine,and glycine might be concerned with tumor cell differentiated degree.Proline and glycine might be important pathologic typing index.It was also concluded that alanine was concerned with axillary ??h nodes metastasis.Serine,threonine, aspartic acid and glycine were concerned with tumor growth. Leucine, valine, isoleucine, histidine and tyrosine might keep body weight in patients with breast cancer.
