Objective To observe protective effects of Dendrobium nobile lindl on liver and behavior of rat with chronic fatigue syndrome, so as to provide reference data for the development and application of DNL and prevention of CSF.Methods Female SD rats were randomly divided into 7 groups of 11 and received different treatments, including negative control group(group N), positive control group(group P1, group P2), the flower of DNL intervention group(group DF), DNL intervention group(including group DL, DM, DH). CFS model was created by swim exhaustion and sleep deprivation. When the model was established, group P2 was sacrificed for collecting tissues such as liver, blood. Group DF was treated by gavage of different agentia described above once a day, such as DNL flower and three concentrations of DNL, named as group DL, DM and DH(0.1g/100 g.bw, 0.3g/100 g.bw and 0.5g/100 g.bw) respectively, while group N and group P1 were given isovolumetric distilled water. After fourteen days, Morris water maze(MWM) was performed to evaluate the learning-memory ability of rat. In the end, the rats were sacrificed to collect liver and blood for further detection.Results According to the observation of ethological indexes, the sleep deprivation model was successfully built in this experiment. In Morris water maze(MWM), group P1 spent longer time for finding the platform than group N(P<0.05), but group DL, DM, DH, DF spent shorter time than group P1(P<0.05); the result of space exploration test showed group P1 crossed less times and moved shorter distance than group N(P<0.05), but the times and distance increased among group DL, DM, DH, DF(vs. group P1, P<0.05). According to the results of serum SOD activities, the serum SOD activities of group P1 decreased markedly when compared with group N, but it increased when intervening withDNL and its flower(P<0.05). Besides, according to the results, the content of serum MDA, AST and ALT increased significantly in group P1(vs. group N, P<0.05), but it decreased markedly among group DL, DM, DH compared with group P1(P<0.05). What’s more, hepatic histopathological observation showed cell edema, congestion, fibrosis and inflammatory cell appeared in liver tissue of group P1, while hepatic inflammatory was milder among group DL, DM, DH, DF compared with group P1, and improved with increase of dosage, besides, none of significant difference among group DH, DF and N was not found.Conclusion DNL can effectively improve the behavior and physical status of CSF rats, the effects, for which, removing free radicals may be one of underlying anti-fatigue mechanisms of DNL. The effect to enhance learning-memory of DNL flower was better than that of DNL. And both of them have protective function to the liver of rat with CFS.
