Objectives:Silkworm allergy is a kind of with Chinese characteristics of management ofoccupational allergy and a food allergy,silkworm protein has strong allergen activityis the main allergens.Establish a silkworm chrysalis coarse extraction immersionsensitization BALB/C mice intestinal and asthma model; Detection of specific IgElevels and IgG, spleen cell factor changes, gut and lungs HE staining pathologyanalysis. This research has a distinct Chinese characteristics, lay a foundation for thediagnosis and treatment of silkworm allergy.Methods:1. The silkworm chrysalis coarse extraction BALB/C mice intestinal allergicsensitization to immersion method of model:Will only325-6weeks close female BALB/c mice were randomly divided intogroup (PBS) negative group, positive control group the cholera toxin (CT), silkwormchrysalis coarse immersion group (SPE), cholera toxin+silkworm chrysalis coarseextraction immersion group (CT+SPE group)8in each group. Negative groupslavage PBS; Immersion in positive lavage silkworm chrysalis coarse, interval of3days, lavage6times. Lavage inspire3times, lavage anal temperature before and afterthe test, observe sensitization state; Stimulate gut after sensitization model; Collectserum in mice, and collect the mice spleen cells supernatant, measure: sIgE, sIgG2a,cytokines (IFN-gamma, IL-5, IL-4). Jejunal segment tissue pathological changesin mice and other indicators to evaluate the construction of a model.2. Silkworm chrysalis coarse extraction immersion sensitization asthma model inmice methods:Will be16of5-6weeks close female BALB/c mice were randomly divided intonegative group group (PBS), and the positive group (SPE)8in each group. Negativegroups abdominal subcutaneous injection of PBS; The positive peritoneal injection ofsilkworm chrysalis coarse immersion sensitization,3times in a row, two weeks at a time. Intranasal and inspire a day once a week. Airway hyperresponsiveness afterexcitation test; Collect BALF (alveolar lavage) cell count, classification.24hours tocollect serum in mice; Detecting specific antibody (IgE), cytokines (IL-4, IL-10,the INF-gamma) and lung tissue pathological changes such as index to evaluate theconstruction of a model.Results:1. The silkworm chrysalis coarse extraction BALB/C mice intestinal allergicsensitization to immersion model results:BALB/c mice CT+SPE group anal temperature change, allergic to state theobvious; Silkworm coarse immersion intestinal tissue pathological changes, there wasan obvious inflammatory lesions; Spleen cell culture supernatant on IL-4, IL-5,INF-gamma reduce; Eosinophil increased (P <0.05), no negative group, all theindicators.2. Silkworm chrysalis coarse extraction immersion sensitization BALB/C miceasthma model results:Silkworm coarse immersion group of lung pathological changes, there was anobvious inflammatory lesions, BALF cell count increased alveolar lavage fluid,lavage IL-4, IL-10, INF-gamma reduce; Eosinophil increased (P <0.05), nonegative group, all the indicators.Conclusion:Silkworm chrysalis coarse extraction immersion lavage methods BALB/C miceintestinal allergic sensitization to modeling success; Silkworm chrysalis coarseextraction immersion sensitization BALB/C mice allergic asthma model success. Twoallergy model lays the foundation for later silkworm chrysalis allergy diagnosistreatment.
