【Background】Heat shock protein70(HSP70) is a kind of highly conserved proteins which isgenerated in the stress state. There is HSP70expressive in many tumors. HSP70playsits anti-tumor role mainly by affecting the cell apoptosis. Helicobacter pylori (Hp)chronic infection in gastric cancer plays a promoting role in the development processof gastric cancer, it may involve in the occurrence of gastric cancer by promotingtumor-associated genes such as p53gene in gastric epithelial cells mutate. Mutant p53(mtp53) plays an important role in the process of gastric cancer development.However, it has not been researched extensively that expression of HSP70, p53inGastric Carcinoma and their Relationship with HP Infection. Analyzing expression ofHSP70and p53in gastric cancer tissue and their relationship with HP infection, canprovide a new train of thought in the mechanism of HP in stomach cancer. It can alsoprovide a certain theoretical basis for the early diagnosis of gastric cancer and thejudgment of gastric cancer’s malignant degree.【Objectives】1. Understand the expression of HSP70and p53in gastric cancer tissue.2. Explore the relationship between HSP70, p53expression andclinicopathological features in human gastric carcinoma.3. Analysis of whether there is a correlation between the expression of HSP70and the expression of p53in gastric cancer.4. Discuss whether there is a correlation the expression of HSP70and p53ingastric cancer with HP infection. 5. Analysis of the possible mechanism of HSP70, p53and HP infection in thedevelopment of gastric cancer comprehensively.【Methods】1. A total of56cases of gastric adenocarcinoma tissues were selected fromYanan University Affiliated Hospital in May2012-May2013. These cases wereresected by surgical and were confirmed by pathological diagnosis. Meantime, theyhave not been given by radiotherapy, chemotherapy and other anti-cancer treatmentbefore surgical. Select12cases of normal gastric mucosa tissues as control group. Hpinfection was detected by Warthin-Starry stain in gastric cancer group. The expressionof HSP70and p53in all these cases was detected by SABC Immunohistochemicalmethod.2. Collect the information of the selected patients, including the generalcharacteristics of selected cases, such as demographic characteristics, clinical andpathological features.3. Statistical analysis: The data was processed by SPSS17.0statistical software.The χ2test or Fisher’s exact test was used to assess the significant difference betweenthe groups. Spearsman correlation analysis was used to assess the significantdifference between the variable datas. P<0.05is considered to have statisticallysignificant.【Results】1. The positive expression rate of HSP70in gastric cancer tissue was82.14%(46/56), the positive expression rate in normal gastric tissues was33.33%(4/12).There was statistically significant between them (P <0.01). The positive expressionrate of HSP70in gastric carcinoma had no correlation with the patient’s gender, ageand tumor location (P>0.05). The positive expression rate of HSP70in gastriccarcinoma was significantly correlated with the degree of differentiation of gastriccarcinoma, lymph node metastasis, distant metastasis and clinical stage (P <0.05). 2. The positive expression rate of the mutant p53in gastric cancer tissue was62.50%(35/56), there was no positive expression in normal gastric tissues (0/12).There was statistically significant between them (P <0.01). The positive expressionrate of p53in gastric carcinoma had no correlation with the patient’s gender, age,tumor location, degree of differentiation and distant metastasis (P>0.05). Positiveexpression rate of p53in gastric carcinoma was correlated with lymph nodemetastasis, clinical stage and gastric cancer (P <0.05).3. In the detection of HP infection in gastric cancer tissues, there were32caseswith positive HP infection(57.14%) and24cases with negative HP infection(42.86%).4. In the56cases of gastric cancer tissues, the positive expression rate of HSP70in gastric carcinoma group with positive HP infection was93.75%(30/32), thepositive expression rate of HSP70in gastric carcinoma group with negative HPinfection group was66.67%(16/24). There was statistically significant between thetwo groups (P <0.05). In the56cases of gastric cancer tissues, the positive expressionrate of p53in gastric carcinoma group with positive HP infection was87.50%(28/32),the positive expression rate of p53in gastric carcinoma group with negative HPinfection group was29.17%(7/24). There was statistically significant between thetwo groups (P <0.05).5. In the56cases of gastric cancer tissues, the conjunct positive expression rateof HSP70and p53is57.14%(32/56). The positive expression of HSP70wascorrelated with the positive expression of p53in gastric cancer (P <0.05).【Conclusions】1. The positive expression of HSP70in gastric cancer tissue was obviouslyhigher than that of normal tissue. The positive expression of HSP70was correlatedwith the degree of differentiation, lymph node metastasis, distant metastasis andclinical stage. Detecting the expression of HSP70in gastric cancer tissue can providea certain basis for the malignant degree and prognosis of gastric cancer. 2. There was abnormal expression in gastric cancer tissue of p53. The positiveexpression of p53in gastric cancer was correlated with degree of differentiation,lymph node metastasis and clinical stage. It was closely related with the incidence andthe development of gastric cancer.3. In the56cases of gastric cancer tissues, the positive expression of HSP70inpositive HP infection group was obviously higher than that of negative HP infectiongroup, the positive expression of p53in positive HP infection group was obviouslyhigher than that of negative HP infection. HP infection may be one of the precipitatingfactors in the high expression of HSP70and mutant p53.4. The expression of HSP70had a certain correlation with the expression of p53,they may participate in the occurrence of gastric cancer development commonly.
